| Yellow Camellias is a plant of the genus Camellia in the Theaceae family that was identified in the early 1960s and is primarily located in China’s Guangxi Zhuang Autonomous Region.It is one of China’s first-class protected species.Yellow Camellias has pharmacological properties such as antitumor,antioxidant,hypolipidemic,hypoglycemic,antibacterial,and so on.Its chemical constituents is complex,and the currently reported chemical constituents include flavonoids,saponins,phenylpropanoids,volatile substances,and other chemical constituents.There are many varieties of yellow Camellias that are confusingly circulated in the market,and the composition and content of substances about different varieties and parts of yellow Camellias are still not perfect.As a result,an analytical method is urgently needed to provide the theoretical basis for quality control,as well as further development and utilization of yellow Camellias.The purpose of this study is to develop a sensitive,quick,and dependable analytical method for quality control,composition identification,and content determination of yellow Camellias using liquid chromatography-mass spectrometry(LC-MS).In this paper,the leaves and flowers of five species of yellow Camellias were studied for their systematic chemical constituents analysis based on liquid chromatography-mass spectrometry.The following are the primary research findings:(1)An analytical method based on UHPLC-QTRAP-MS/MS technique was developed for the rapid identification of chemical constituents in yellow Camellias.In this study,138 chemical components were identified from the alcoholic extracts of leaves and flowers of five varieties of yellow Camellias,including 85 flavonoids,14 phenylpropanoids,22 phenolic acids,9 terpenes and their saponins,and 8 others(6 fatty acids and 2 sterols).In addition,14compounds among these 138 chemical components were identified for the first time in yellow Camellias by liquid chromatography-mass spectrometry.(2)Using the multiple reaction monitoring mode(MRM)of the UHPLC-Qq Q-MS/MS technique,a method was devised for the simultaneous measurement of 16 chemical components in yellow Camellias.The peak areas and concentrations of the 16 chemical components in yellow Camellias had good linearity(r~2=0.9955~0.9998),intra-Day precision(RSD=0.89%~2.76%),inter-Day precision(RSD=0.96%~2.51%),repeatability(RSD=2.11%~8.08%),stability(RSD=1.24%~9.74%),and spiked recovery was 95.58%~103.41%according to the methodological validation.The method is accurate,quick,and simple,and it can be used to determine the content of yellow Camellias.The results indicated that there were differences in the contents of compounds in the leaves and flowers of five species of yellow Camellias.The quantitative approach developed in this study can offer a trustworthy analytical tool for yellow Camellias quality control.(3)The LC-MS fingerprint profiles of C.nitidissima from different picking months were established,the shared peaks were tagged and identified,and the diverse batches of C.nitidissima were categorised by combining chemometric approaches.Furthermore,LC-MS fingerprint profiles for flowers and leaves of different yellow Camellias varieties were produced,and the level of variability among variations was assessed using the similarity evaluation system for the chromatographic fingerprint of Traditional Chinese Medicine software.This study lays a good foundation for the comprehensive evaluation and quality control of yellow Camellias,which is of great significance for the quality control of different yellow Camellias varieties and medicinal parts in terms of identification and content determination. |
