| Objective: In this study,the expression of Epstein-Barr virus-encoded RNA(EBER),programmed death-ligand 1(PD-L1),CD4 and CD8 in diffuse large B-cell lymphoma(DLBCL)and Hodgkin lymphoma(HL)was detected,and their relationship and significance were probed.Methods: A total number of 70 paraffin tissue samples from patients diagnosed with DLBCL and HL were collected,including 55 cases of DLBCL and 15 cases of HL.The expression of EBER was detected by in situ hybridization(ISH)and the expression of PD-L1,CD4 and CD8 was detected by immunohistochemistry(IHC)staining in DLBCL and HL.Statistical analysis software SPSS 26.0 was used for statistical analysis of the data.In DLBCL,Nonparametric tests Mann-Whitney U test was used to analyze the expression differences of PD-L1,CD4 and CD8 in different clinicopathological characteristics.Spearman Correlation Analysis was used to analyze the relationship between PD-L1 and CD4 and CD8,the test level α=0.05,P<0.05 was considered statistically significant.Results: 1.Among 55 cases of DLBCL,EBER positive cells were detected in 17 cases(30.9%),while 5 cases(9.1%)reached the positive criteria.Among 15 cases of HL,EBER positive cells were detected in 10cases(66.7%),while 6 cases(40.0%)reached the positive criteria.2.The positive rate of PD-L1 in DLBCL was 72.7%(40/55),including 17 cases with low expression,12 cases with medium expression and 11 cases with high expression.There was no significant difference in the expression of PD-L1 between different genders,ages,inspection parts and pathological classifications of patients(P>0.05).The positive rate of PD-L1 in HL was 80.0%(12/15).3.PD-L1 expression was positive in all 5 EBER positive cases of DLBCL,including 1 case with medium expression and 4 cases with high expression.4.In DLBCL,T lymphocytes infiltrating in the tumor area expressed CD4,including 19 cases with low expression,27 cases with medium expression and 9 cases with high expression.While CD8 expression included low expression in 28 cases,medium expression in 23 cases and high expression in 4 cases.There was no significant difference in the expression of CD4 and CD8 between different genders,ages,inspection parts and pathological classifications of patients(P>0.05).In HL tissues,all infiltrated T lymphocytes had high expression of CD4,9 cases had low and medium CD8 expression,and 6 cases had high CD8 expression.5.In DLBCL,the expression of PD-L1 was positively correlated with CD4 and CD8(P<0.05).Conclusion: 1.The expression of EBER is closely related to HL,and is one of diagnostic bases for HL.EBV infection may be one of pathogenic factors.2.PD-L1 was positively correlated with the expression of CD4 and CD8 in DLBCL,which may provide a theoretical basis for immunotherapy in patients with DLBCL.3.In this study,PD-L1 expression was high in most of the 5 cases of DLBCL with EBER positive,suggesting that EBV infection may affect the expression of PD-L1.However,due to the small number of EBER positive cases,further correlation studies need to be conducted on the basis of larger sample number. |
