Objective: In this study,a rat model of pneumonia was prepared by intranasal instillation of lipopolysaccharide(LPS).By observing the effects of Mongolian medicine Erihemu-8powder on the general condition,body temperature,lung index,lung pathological changes of LPS-induced pneumonia model rats,and the blood routine related inflammatory indexes and serum IL-6,IL-1β,TNF-α,CRP and PCT levels of pneumonia rats,the therapeutic effect of Mongolian medicine Erihemu-8 powder on pneumonia rats was discussed.Methods: The rat pneumonia model was established by smoking and LPS nasal drip.72 SPF male SD rats were randomly divided into six groups : normal group,model group,positive group(Pudilan group),low,medium and high dose group of Erihemu-8,12 rats in each group.Adaptive feeding for a week.Except for the normal group,the rats in the other groups were smoked for10 days to establish a rat pneumonia model by LPS nasal drip.The treatment was started 12 hours after modeling.The normal group and the model control group were given normal saline,the Pudilan group was given Pudilan oral liquid 3.5 g/kg,and the Mongolian medicine Erihemu-8 low,medium and high dose groups were given 0.14 g/kg,0.27 g/kg and 0.54 g/kg,respectively.Intragastric administration for 10 days.The general condition and body temperature of rats were observed during the period.After 12 hours of fasting water after the last administration,the rats were anesthetized with ether.After the rats were completely anesthetized,blood was collected from the abdominal aorta,and some whole blood was taken to detect the levels of WBC,NE,NE%,LY% and PLT.Another part of the whole blood was centrifuged at low temperature(-4 °C,3500 r/min,centrifuged for 10 min),and the supernatant was taken.The levels of serum IL-6,IL-1β,TNF-α,CRP and PCT were detected by ELISA.The complete lung tissue was taken out,and the color and shape of the lung were observed by the naked eye.The lung index was measured by weighing method.The middle lobe of the right lung was taken and HE staining was performed to observe the pathological changes of the lung tissue.Results:1.General situation : The rats in the normal group were in good condition,normal activity,bright fur,normal breathing and normal diet.The rats in the model group had runny nose,reduced activity,weight loss,shortness of breath,dark hair and erect hair after modeling.Eating water intake plummeted.The nose of some rats was red,and pink secretions appeared in the nose of severe cases.After 10 days of treatment,compared with the model group,the overall situation of different administration groups was better than that of the model group,especially the middle and high dose groups of Erihemu-8,with more activities and more sensitive reactions.2.Changes in body temperature : The body temperature of rats in the model group was significantly higher than that of the normal control group 2~4 days after modeling(P<0.01),and there was no significant difference between the drug group and the model group(P>0.05).3.Changes in body weight : Compared with the normal group,the body weight of the model group decreased significantly(P < 0.01).During the administration period,the body weight of rats in the Pudilan group,Erihemu-8 low,medium and high dose groups was higher than that in the model group(P < 0.01).4.Changes in lung index : Compared with the normal group,the lung index of the model group was significantly increased(P < 0.01).Compared with the model group,the lung index of Pudilan group,Erihemu-8 low,medium and high dose groups decreased significantly(P < 0.01).5.Changes of WBC,NE,NE %,LY % and PLT in blood routine of rats : Compared with the normal control group,the levels of WBC,NE and NE % in the blood of the model group were significantly increased(P < 0.01),and the levels of LY % were significantly decreased(P <0.01).Compared with the model group,the levels of WBC,NE and NE % in Pudilan group,Erihemu-8 low,medium and high dose groups decreased to varying degrees(P < 0.05),and the levels of LY % and PLT increased significantly(P < 0.05).6.Analysis of the results of the determination of TNF-a,IL-1β,IL-6,CRP and PCT in serum of rats : Compared with the normal group,the levels of IL-6,IL-1β,TNF-α,CRP and PCT in the model group were significantly increased(P < 0.01).Compared with the model group,the levels of serum IL-6,IL-1β,PCT,TNF-α and CRP in the Pudilan group and the low,medium and high dose groups of Erihemu-8 were decreased to varying degrees(P < 0.01 or P< 0.05).7.Pathological observation of lung tissue in each group :(1)Observation of lung morphology : Compared with the normal control group,the lung color of the model group was dark red,the swelling was more serious,and the lung lobes were not easy to be completely separated.Compared with the model group,the lung color of the rats in the middle and high dose groups of Erihemu-8 was lighter,there was no obvious swelling,the lung lobes could be completely separated,and the elasticity was good.(2)Observation of HE pathological sections of lung tissue : There was no abnormality in the lung tissue of the normal group.The lung tissue structure of the rats in the model group was severely abnormal,the alveolar arrangement in the field of vision was disordered,the alveolar boundary was blurred,a large number of alveolar epithelial cells proliferated,the alveolar atrophy was accompanied by thickening of the alveolar wall,a large number of inflammatory exudation was seen in the bronchus,and a small amount of inflammatory cell infiltration was seen in the tissue.Compared with the model group,the pathological changes of lung tissue in the Pudilan group,the low,medium and high dose groups of Erihemu-8 were improved to varying degrees,especially in the high dose group of Erihemu-8.Conclusion:1.Erihemu-8 powder has a therapeutic effect on LPS-induced pneumonia model rats at an effective dose.2.Erihemu-8 powder can effectively reduce the inflammation of lung tissue in LPS-induced pneumonia model rats.3.Erihemu-8 powder has a regulatory effect on related cytokines in LPS-induced pneumonia model rats.
