Objective:The anti-inflammatory and antipyretic effects of Mongolian Sanchen pill on LPS-induced acute pneumonia model rats were preliminarily discussed,which was the basis for clinical experiments.Methods:1.Study on the antipyretic and anti-inflammatory effects in LPS-induced pneumonia model rats.72 SD rats were randomly divided into blank group,model group,positive group(Qingkailing group),and 12 cases of sanchen pill(low,medium and high)dose group in 6 groups.The treatment effect of each group was observed.In blank control group and model group,0.5%CMC-Na suspension;aqueous solution(0.27 g / Kg)with 0.5%CMC-Na suspension solution at 20 ml/kg;in Sanchen pill group,Sanchen pill(low,medium and high)(0.45 g / Kg,0.9 g / Kg and 1.35 g / Kg)with 0.5%CMC-Na suspension at 20 ml/kg for 14 d once a day.On day 15,all rats received lipopolysaccharide(LPS)at 2mg / kg.2.Rats in each group drank free water,gavage for 2 weeks,and the dose was updated according to the weight change for 2 days.3.The body temperature of rats in each group was measured twice after mold making,at 3 hours and 5 hours after mold making,respectively.The hair,activity and clinical symptoms of each group were observed.Specimens were collected at 8 hours after mold making.By ELISA),anti-inflammatory medium release of serum IL-10,TNF-a,CRP and MCP-1,and HE staining of lung tissue to evaluate the antipyretic,anti-inflammatory and protective effects of Sanchen pill on the acute pneumonia model.Results:1.Rats in the normal control group had normal clinical signs,normal breathing sounds and sensitive response;rats in the model group had less responsive,reduced activity,and accelerated breathing.Compared with the model group,the overall condition of each dose group was better,especially the high dose group was more active and more sensitive.2.The average daily weight gain of rats in each group was not significant(P> 0.05);3.Compared with the blank group,the body temperature of the model group increased after the injection of LPS into the trachea,showing a statistically significant difference(P <0.05).Compared with the model group,the body temperature decrease was statistically significant(P <0.05).4.Compared with the normal control group,the serum TNF-a,MCP-1 and CRP content was increased significantly(P <0.01),while the serum IL-10 content was significantly decreased(P <0.01);indicating successful molding.Compared with the model group,the treatment group(three dose groups and Qingkaeling group)could reduce the inflammatory factors TNF-a,MCP-1 and CRP to different degrees,and the content of IL-10 increased,among which the test indexes of the group were significantly different(P<0.01).All the test indexes of sanchen pill high dose group were better than those of low and medium dose groups;5.HE staining: Compared with the blank group,the lung tissue showed large infiltration of inflammatory cells and alveolar stromal thickening in the model group.Compared with the model group,pulmonary inflammatory exudation and alveolar septal thickening were significantly reduced in the treated group.Conclusion:1.Mongolian sanchen pill can reduce the exudation of inflammatory cells by the expression of TNF-A,CRP,IL-10 and MCP-1 in the serum of LPS-induced pneumonia model,and can effectively reduce the pathological changes of rat lung tissue,thus playing an anti-inflammatory and antipyretic role.2.Mongolian Sanchen pill may be involved in regulating the release of inflammatory mediators,antipyretic and reducing lung injury in pneumonia model rats. |
