Objective: To systematically explain the research progress of "Gall Disease" in the past 20 years,standardize the processing parameters of Baifupian,one of the toxic traditional Chinese medicine processed from Aconite in the empirical formula for treating "Gall Disease",and preliminarily explore the material basis and mechanism of action of Xingqi Xiaoying pellet.Methods: Through the information visualization technology CiteSpace,we constructed and analyzed the knowledge map of "gall disease" literature in the past two decades,and systematically analyzed its research status,hot spots and frontiers;Using cooking time,soaking and bleaching times,and steaming time as evaluation factors,and using single ester alkaloid content,double ester alkaloid content,ash residue,and bile residue as evaluation indicators,the processing parameters of Baifupian were optimized using the Analytic Hierarchy Process(AHP)-Coefficient of Variation TOPSIS model;Based on interdisciplinary technologies such as modern analytical technology,network pharmacology,molecular docking,and cell biology,this study systematically elucidates the characteristics of Xingqi Xiaoying pellet’ multi component,multi target,and multi pathway intervention in "gall disease".Results:(1)CiteSpace knowledge graph visualization analysis: Previous research has mainly focused on diagnostic stratification,cancer,surgery,and ablation;Association guidelines,ablation,and diagnostic stratification may be hot topics in the study of "gall disease".The dialectical analysis,clinical efficacy,and mechanism of action of traditional Chinese medicine in the treatment of different types of "gall diseases" are the research trends of traditional Chinese medicine in the treatment of "gall diseases".There is relatively little research on the therapeutic drugs and their mechanisms of action for "gall disease".(2)The optimal processing technology for Baifupian is: soak in bile: remove the fibrous roots of fresh aconite,place it in a bile pool for several days,add 30 kg of bile to every 100 kg of aconite,and add water.Processing: Take out the monkshood and boil it with the soaking solution for 20 minutes.Take it out,let it cool,peel it off,and cut it into 3mm thin slices.Soak in clean water for 2 days,change water 3 times a day,steam for 3 hours,and dry in the sun.(3)Based on UPLC-Q-TOF-MS technology,93 compounds in Qixiaoying pellet were identified;Based on network pharmacology,there are 9 main pathways that predict the mechanism of Xingqi Xiaoying Granule in treating "gall disease",namely:cell apoptosis signaling pathway,PI3K-Akt signaling pathway,JAK-STAT signaling pathway,MAPK signaling pathway,etc.Key targets include AKT1,BAX,BCL2,CASP3,TNF;The six key compounds are: kaempferol,ursolic acid,saikosaponin A,luteolin,isoliquiritigenin,and rutin.Molecular docking experiments showed that the binding of 6 key compounds to 5 key targets was relatively stable.In vitro cell experiments have shown that rutin,ursolic acid,kaempferol,and luteolin have significant inhibitory effects on the growth of human thyroid cells Nthy ori 3-1.The inhibitory effects of Chaihu saponin A and isoliquiritigenin on the growth of Nthy ori3-1 cells were not significant.Conclusions: This study systematically reviews the research progress of "Gall Disease" in the past 20 years,standardizes the processing parameters of Baifupian,one of the toxic traditional Chinese medicine processed products in the empirical formula for treating "Gall Disease",explores the material basis and mechanism of action of Xingqi Xiaoying pellet,provides reference for the clinical treatment of "Gall Disease",and also provides reference for the processing of other medicinal materials,the pharmacological material basis and mechanism of action of traditional Chinese medicine formulas and preparations.
