Effect Of Qiyu Sanlong Tang Containing Serum Mediating Per2 Regulation Of P53-Mdm2 Axis On The Proliferation Of A549 Cells

Objective: To study the influence of QYSLT on biological clock gene Period2(Per2)to inhibit A549 cell proliferation,and the influence of Qys LT medicinal serum on the expression of biological clock gene Per2 through the regulation effect of Per2 on P53-Mdm2 axis.Methods: Human A549 cells were selected as the target,and the effect of A549 cells synchronized with Dexamethasone(DEX)on the proliferation of A549 cells was examined by the Cell Proliferation Assay Kit(CCK-8).Plate cloning experiment demonstrated the effect of Qi-Yusan-long decoction medicated serum with gradient concentration on the proliferation of A549 cells.The effect of QYSLT drug-containing serum on cell cycle of A549 was detected by Flow Cytometry(FCM),and cell proliferation was observed by SPF and PI values.Real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)was used to first screen the best small interference sequences from hs-per2-si1,hs-per2-si2 and hs-per2-si3,and then detect the m RNA expression of Per2,P53 and Mdm2 under the influence of Qiyusanlong decoction.The protein expression of Per2,P53 and Mdm2 was determined by Western blot.After the intervention of si-Per2 gene infection,the expressions of si-Per2,P53 and Mdm2 in m RNA and protein levels were detected by RT-qPCR and Western blot.Results: 1 Qi-Yusan-long Decoction medicated serum can inhibit the cell activity best at 20% concentration for 24 h 2 Compared with blank serum group,Qi-Yusan-long Decoction medicated serum group can effectively inhibit the proliferation of A549 cells(P<0.01)3 Compared with blank serum group,The expressions of Mdm2 m RNA and protein in A549 cells were decreased(P<0.01),while the expression levels of Per2,P53,m RNA and protein were increased(P<0.01).Compared with the blank control group,A549 cells cultured in the medicated serum group of Qiyusanlong Decoction effectively inhibited cell proliferation.Conclusion: Qiyu Sanlong Decoction can promote the expression of Per2 and P53 m RNA and protein in A549 cells of lung cancer,reduce the expression of Mdm2 m RNA and protein,induce cell cycle arrest of lung cancer,and inhibit tumor proliferation.
