Aim:The effect of 5-FU on liver cancer mice was treated with EA to clarify the pathological rhythm of spontaneous activity and the effect of elective EA on the rhythm of liver cancer mice,and to study the mechanism of elective EA on Per gene in SCN and peripheral tissues of liver cancer chemotherapy in mice.Method:Thirty-five C57 BL / 6J 6-7 weeks old male mice were divided into blank group,model group,EA group,chemotherapy group and EA+ chemotherapy group,each group of seven.Then,the mice were acclimated for 10 days after the automatic light-dark control(L:D was 12: 12,07: 00-19: 00 for the light period,19:00-the next day 07:00 for the dark),then the H22 liver cancer cell line was injected into the mouse liver to establish the orthotopic liver transplantation model.Mice were subjected to experimental intervention after successful modeling,EA group was given EA intervention;the chemotherapy group received 5-FU intraperitoneal injection;Chemotherapy + EA group given 5-FU intraperitoneal injection immediately after the start of electro-acupuncture intervention.EA method: select the "gan shu point" and "zhi yang point" 2 for EA.EA set: 2/15 Hz,0.5m A,each time 15 min,once a day,a total of 7 times.Animals were sacrificed with the tenth electro-acupuncture after 2h,and their SCN and peripheral tissues were harvested.Before and after the modeling,Export the data of Biological rhythm before and after the modeling and intervention,then analyzed it,and the expression of SCN and Peripheral tissue Per1 and Per2 gene was detected by RT-PCR after chemotherapy and EA.Result:(1)There was no significant difference in the start time,cycle,amplitude and peak phase of circadian rhythm of spontaneous activity among the animals(P>0.05).The spontaneous activity was similar to that of circadian rhythm.(2)Compared with the blank group,there were significant differences(P<0.05)in the model group,and the difference between the amplitude and the phase shift of the model group was statistically different(P<0.05).(3)After chemotherapy,the animals in the chemotherapy group were delayed than those in the model group,and there was no significant difference in the start time,amplitude and period difference between the two groups before and after chemotherapy(P>0.05).(4)Chemotherapy + EA,chemotherapy + EA group spontaneous activity of circadian rhythm delayed phase forward,there was no significant difference in the start time,amplitude and period difference between the two groups before and after chemotherapy +EA(P>0.05).(5)After chemotherapy,the expression of Per1 and Per2 in the SCN and peripheral tissues of the chemotherapy group were lower than those in the model group,and the differences were statistically significant(P<0.05).The expression of Per1 and Per2 in the SCN and peripheral tissues of chemotherapy group and electroacupuncture group was higher than that of chemotherapy group,but there was no significant difference between the two groups(P>0.05)Compared with the model group,Per1 and Per2 were down-regulated in SCN and peripheral tissues,and the differences were statistically significant(P<0.05).Conclusion:(1)The circadian rhythm of spontaneous activity was changed to some extent after C57 BL / 6J mice were transplanted into liver cancer.The main activity was reduced and the total activity decreased and the phase lag was not significant change.(2)In the LD operating state,chemotherapy can adjust the circadian rhythm of spontaneous activity of liver cancer to a certain extent,so that the delayed phase is shifted forward,and its initial time,period and amplitude have no obvious adjustment effect.(3)In the LD operating state,the electroacupuncture can adjust the circadian rhythm of spontaneous activity of the liver cancer mice to a certain extent,so that the reduced amplitude is increased and the delayed phase is advanced,but the circadian rhythm of the spontaneous activity of the liver cancer mice after chemotherapy No significant adjustment effect.(4)The regulation of Per gene and other related rhythmic genes in SCN and peripheral tissues of liver cancer may be one of the mechanisms of electroacupuncture to adjust the biological rhythm.
