syndrome classification,clarify the correlation between the patient’s TCM syndrome classification and clinical pathological characteristics;use ELISA to detect 120 non-small cell lung cancer patients The expression levels of Bcl-2 and Bax in the serum of 30 healthy people were analyzed,and the diagnostic significance and value of two apoptotic indicators in blood samples of NSCLC patients were analyzed.2.60 SD rats were selected and randomly divided into a blank control group and a Chinese medicine group,and were administered with 1m L / 100 g physiological saline and 1m L / 100 g Qiyu Sanlong Decoction(raw medicine content: 1.396 g / m L).Blank serum and Qiyu Sanlong Decoction containing medicated serum.A549 is a human lung adenocarcinoma cell.This experiment is based on different groups,adding different concentrations of Qiyu Sanlong Decoction-containing serum and western medicine to interfere with JAK2,STAT3 and apoptosis-related factors Bcl-2,Bax in the JAK2 / STAT3 signaling pathway.Observation and detection were conducted to investigate whether Qiyu Sanlong Decoction can induce apoptosis by blocking JAK2 / STAT3 signaling pathway and then regulating apoptosis-related factors.Results 1.In terms of gender distribution,79 were male(65.8%),41 were female(34.2%),and the male to female ratio was 1.93: 1.In the pathological type distribution,72 were adenocarcinoma(60%),and 45 were squamous cell carcinoma(37.5%).In clinical TNM staging,5 cases were in stage I(4.17%),5 cases in stage II(4.17%),38 cases in stage III(31.7%),and 72 cases in stage IV(60%).Among 120 cases of non-small cell lung cancer,the type of TCM syndromes is mainly Qi deficiency and phlegm dampness,accounting for 43.33%,followed by Qi deficiency and yin deficiency(21.67%),Qi stagnation and blood stasis(15%),and Yin deficiency toxin-heat type.(11.67%)and phlegm heat-resistant lung type(8.33%).2.After statistical analysis,no difference between smoking and TCM syndrome distribution.There was no significant difference in gender composition and age distribution among different syndrome types.NSCLC is mainly lung adenocarcinoma and lung squamous cell carcinoma.According to statistical analysis,the pathological type of NSCLC patients is lung squamous cell carcinoma and lung adenocarcinoma.The medical syndrome types are basically similar.The clinical symptoms are qi deficiency,phlegm and dampness,and qi and yin deficiency.Types are more common.The stage of NSCLC TNM is mainly stage III and stage IV.According to statistical analysis,the TCM syndrome types of patients with stage III and stage IV NSCLC are basically similar.3.The distribution of apoptosis-related factors Bcl-2 in clinical serum samples of NSCLC patients is not significantly different from that of healthy controls.Bax is distributed differently between the two groups.Bax in serum samples can be used as an auxiliary indicator for clinical diagnosis of NSCLC.4.Qiyu Sanlong Decoction-containing serum inhibits the proliferation of A549 cells in a concentration-and time-dependent manner,with the optimal concentration of 20% and the optimal time for 24 hours;AG490 inhibits the proliferation of A549 cells in a concentration-,time-dependent,and optimal concentration It is 75 umol / L,and the best action time is 24 h.RT-q PCR showed that Qiyu Sanlong Decoction-containing serum could inhibit Bcl-2 m RNA expression and promote Bax m RNA expression,but had no significant effect on the transcription levels of JAK2 m RNA and STAT3 m RNA.Western Blot showed that Qiyu Sanlong Decoction’s serum can inhibit Bcl-2 protein level,promote Bax protein expression,and inhibit p-jak2,p-stat3 protein expression.5.Response experiments showed that IL-6 promoted bcl-2 m RNA and protein expression,inhibited bax m RNA and protein expression,while p-JAK2,p-STAT3 protein expression was also increased.The IL-6 + group can significantly inhibit bcl-2 m RNA level and promote bax m RNA expression;it can significantly inhibit the increase of p-JAK2,p-STAT3 protein expression caused by IL-6,and inhibit the increase of bcl-2 protein expression.conclusion 1.The distribution of traditional Chinese medicine syndrome in patients with NSCLC is mostly qi deficiency and phlegm-dampness.The pathological types of lung adenocarcinoma and lung squamous cell carcinoma showed no significant difference between the pathological types and TCM syndrome types;TNM stages were stage III and stage IV NSCLC patients,and there was no significant difference between the pathological stages and TCM syndrome types.The distribution of apoptosis-related factor Bcl-2 in serum samples of NSCLC patients was not significantly different from that of healthy controls.The expression level of Bax was lower than that of healthy controls.Bax can be used as an auxiliary indicator for clinical diagnosis of NSCLC in serum samples.2.Qiyu Sanlong Decoction can induce apoptosis of A549 cells,and its mechanism is related to inhibiting JAK2/STAT3 signaling pathway,inhibiting downstream Bcl-2 level,and promoting Bax expression. |
