Objective: To investigate the expression of YWHAE and CDC6 in colorectal cancer tissue,analyze the relationship between their expression and clinical pathological characteristics and prognosis of patients,and further analyze the correlation between the two.Method: The Cancer Genome Atlas(TCGA)was used to download the expression profiles and clinical datasets of YWHAE mRNA and CDC6 mRNA in colorectal cancer patients.The relationship between their expression and clinical pathological characteristics and prognosis of colorectal cancer patients was analyzed,and the correlation between the two was also analyzed.Obtain co-expressed genes of CDC6 and YWHAE through the cBio Portal website,and further enrich and analyze the coexpression genes.Collect paraffin tissue samples from colorectal cancer patients in our hospital and make them into tissue chips.Use RNAscope technology to detect the expression of YWHAE mRNA and CDC6 mRNA,analyze their relationship with clinical pathological features,and analyze the correlation between the two.Immunohistochemical methods were used to detect the expression of YWHAE and CDC6 in colorectal cancer tissue.Chi-square tests were used to analyze the relationship between their expression in colorectal cancer tissue and clinical pathological characteristics.Kaplan Meier method was used for univariate analysis and survival curves were plotted.Log rank test was used for survival rate comparison,and COX risk proportional regression model was used for univariate and multivariate analysis.Result: 1.The results of bioinformatics analysis showed that the median expression level of YWHAE mRNA in colorectal cancer tissues(423.7)and the median expression level of normal tissues(330.05)were statistically significant(P<0.05),and the median expression level of CDC6 mRNA in colorectal cancer tissues(22.81)and the median expression level of normal tissues(6.04)were statistically significant(P<0.05).There is a positive correlation between the expression of YWHAE mRNA and CDC6 mRNA in colorectal cancer(P<0.01).The enrichment analysis of co-expressed genes YWHAE and CDC6 showed that both are involved in biological processes such as cell cycle.2.The RNAscope detection results showed that the expression of YWHAE mRNA and CDC6 mRNA in colorectal cancer tissue was significantly higher than that in normal tissue(P<0.05),and the expression of the two in colorectal cancer was positively correlated(P<0.01).3.The immunohistochemical detection results showed that the high expression rate of YWHAE protein in normal tissues was 24.83%,and in cancer tissues was 73.17%.The difference between the two groups was statistically significant(P<0.05);The high expression rate of CDC6 protein in normal tissues was23.49%,and in cancer tissues was 66.46%.The difference between the two groups was statistically significant(P<0.05).The expression of both is related to lymph node metastasis and pathological staging(P<0.05).The expression of both in colorectal cancer is positively correlated(P < 0.01).The average survival time of the high expression group of YWHAE protein and CDC6 protein was significantly lower than that of the low expression group,and the difference was statistically significant(P<0.05),which is related to poor prognosis of patients.Conclusion: 1.YWHAE mRNA and CDC6 mRNA are highly expressed in colorectal cancer tissue and are related to biological processes such as cell cycle.Both may be a molecular event in the early stage of colorectal cancer occurrence.2.YWHAE protein and CDC6 protein are both highly expressed in colorectal cancer tissue,and are associated with lymph node metastasis,pathological staging,and poor prognosis.Both are expected to become biological markers and potential targets for poor prognosis in colorectal cancer patients.3.The expression of YWHAE and CDC6 in colorectal cancer tissue is positively correlated,and they may jointly participate in the occurrence and progression of colorectal cancer,providing a theoretical basis for further research on the molecular mechanisms related to the two.
