ObjectiveYWHAE(14-3-3 ε)is a 30 kDa acidic polypeptide belonging to 14-3-3 protein family,which is called tyrosine 3-monooxygenase / tryptophan 5-monooxygenase activated protein in Chinese.There are seven subtypes of 14-3-3 protein: β,ε,ε,γ,τ,δand σ.YWHAE(14-3-3 ε)is one of them.In eukaryotic cells,studies have shown that the family proteins are highly conserved acidic proteins.By binding with their corresponding target proteins,they participate in and regulate human cell cycle,signal transduction,adhesion and malignant transformation.At present,according to various tumor research directions,YWHAE is closely related to the occurrence and development of a variety of common malignant tumors,such as liver cancer,breast cancer,ovarian cancer,gastric cancer,renal cancer and nervous system tumors,which are all highly expressed in normal tissues.However,there are few reports on the expression of YWHAE in colorectal tumors.In this paper,the expression level of ywhae protein in colorectal cancer tissues and its correlation with the prognosis of patients were studied to explore its clinical significance,and provide possible direction and ideas for the treatment of colorectal cancer in the future.MethodsIn this study,we collected 294 postoperative pathological specimens of colorectal cancer patients in the Department of general surgery,Affiliated Central Hospital of Shenyang Medical College from 2009 to 2014.None of the patients had received preoperative neoadjuvant chemoradiotherapy and had signed the informed consent.Among 294 cases,254 cases were matched successfully between cancer tissue and paracancerous tissue.The prepared tissue microarray was used to detect the location,distribution and expression of YWHAE in colorectal cancer tissues and adjacent tissues by immunohistochemical staining(PV two-step method).Then the data were statistically analyzed to analyze the correlation between ywhae expression and clinical data and prognosis of colorectal cancer patients.ResultsAccording to the results of experimental immunohistochemical staining(PV twostep method),the positive expression of YWHAE was mainly located in the cytoplasm of the cells,and a small amount in the cytoplasm and cell membrane.The high expression rate of YWHAE in colorectal tumor tissues was significantly higher than that in the adjacent tissues(P < 0.05).The positive expression of YWHAE was closely related to the pathogenesis of colorectal cancer The positive expression of YWHAE was positively correlated with histological grade,depth of invasion,lymph node metastasis and TNM stage(P < 0.05);the positive expression of YWHAE was not significantly correlated with gender,age,tumor location and tumor gross classification of colorectal cancer patients(P > 0.05);Kaplan Meier analysis showed that the positive expression of YWHAE was positively correlated with TNM stage(P < 0.05)Results the survival time of patients with high expression of YWHAE was significantly shorter than that of patients with low expression of YWHAE(P < 0.05).Cox regression analysis showed that ywhae was an independent prognostic factor for colorectal cancer patients(P < 0.05).Conclusions(1)YWHAE is highly expressed in colorectal cancer.(2)The positive expression of YWHAE was positively correlated with histological grade,depth of invasion,lymph node metastasis and TNM stage.(3)YWHAE is an independent prognostic factor for colorectal cancer.YWHAE is a potential new target for the treatment of colorectal cancer.
