Objective To investigate the expression of miR-490-3p in gastric cancer,and explore the relationship between miR-490-3p expression level and clinicopathological characteristics,prognosis of gastric cancer,with the aim of providing potential markers for diagnosis and prognostic analysis in patients with gastric cancer.Methods A total of 80 patients undergoing radical surgery for gastric cancer in our hospital from January 201 7 to December 2019 were selected to detect the relative expression of miR-490-3p in cancer tissues and adjacent tissues of gastric cancer patients using real-time fluorescence quantitativ e PCR(qPCR).The relationship between miR-490-3p and tumor markers and clinical pathological parameters of gastric cancer was analyzed.At the same time,Kaplan-Meier curve,log-rank method and Cox proportional risk regression model were used for prognostic analysis.Results The relative expression of miR-490-3p in cancer tissue was0.652±0.072,significantly lower than the expression of 1.259±0.069 in normal tissues adjacent to cancer,with a statistically significant difference between the two(t=40.528,P<0.05);The correlation coefficients between miR-490-3p expression and tum or markers CEA and CA19-9 in gastric cancer tissue were-0.438 and-0.421 respectively(P<0.01);At the same time,according to the mean expression of miR-490-3p,80 patients with gastric cancer were divided into a high expression group of 32 cases and a low expression group of 48 cases.miR-490-3p was not correlated with clinical and pathological characteristics such as gender,age,tumor location,tumor diameter,and degree of differentiation(P>0.05),but was significantly correlated with clinical case p arameters such as CEA,CA19-9,depth of invasion,lymph node metastasis,and TMN stage(χ~2=7.230、7.026、5.275、6.601、6.243,P<0.05);K-M survival curve was used to analyze the impact of miR-490-3p expression on the prognosis of gastric cancer patients.The results showed that the progression free survival in the low expression group of miR-490-3p was significantly lower than that in the high expression group(2 3.0 months vs 30.0 months,P<0.05);Log-rank univariate analysis and Cox multivariate analysis showed that lymph node metastasis,TNM stage,and miR-490-3p were independent risk factors for postoperative survival in patients with gastric cancer(HR=16.747,17.804,and 3.124,all P<0.05).According to the ROC curve anal ysis of the survival of patients with gastric cancer after surgery,the sensitivity and specificity of miR-490-3p in predicting the survival of patients with gastric cancer were 70.8%and 75.0%respectively,and the AUC was 0.712(P<0.05),indicating that miR-490-3p has a certain predictive value for the survival of patients with gastric cancer after surgery.Conclusions The decreased expression of miR-490-3p may promote the proliferation,differentiation,and migration of gastric cancer,and may be a potential molecular marker for evaluating the diagnosis and survival prognosis of gastric cancer.
