Objective:To explore the effect of Tianxing Jianshenwei granule on antioxidant in aging rats induced by D-galactose,and to provide theoretical and experimental basis for further promotion and research of this health care product.Methods:Sixty SPF male SD rats were selected in this experiment,they were randomly divided into blank group,model group,vitamin E group,tianxingjianshenwei granule high-dose,middle-dose and low-dose groups with ten rats in each group.The aging model was induced by intraperitoneal injection of D-galactose.Except the blank group,the other groups were given intraperitoneal injection of D-galactose at a dose of 500 mg/kg/d for six weeks.From the third week,the rats in the blank group and the model group were given the corresponding volume ratio of distilled water by gavage,and those in the vitamin E group were given the corresponding dose of 27 mg/kg/d by gavage,the high,middle and low dose groups of tianxingjianshenwei granule were given by gavage according to the standards of 10.8 g/kg/d,5.4g/kg/d and 2.7 g/kg/d respectively for four weeks.At the end of the last administration,they fasted for 12 hours without stopping drinking.After being weighed and anesthetized,blood was taken from the abdominal aorta,and the liver,kidney,spleen and thymus were taken out completely,and the index of each organ were calculated,the levels of SOD,GSH-Px and MDA in serum,spleen and kidney were measured,and the kidney were detected by HE staining,the changes of tissue morphology were observed.Results:1.The general state of rats: Compared with the blank group,the model group rats’ fur was dry,sparse and dull,and had the phenomenon of depilation.The model group rats moved slowly,languid and prone to lying down,and the skin was loose and lack of elasticity.The general state of vitamin E group and the high,middle and low dose groups of Tianxingjianshenwei granule were improved.2.Organ index of rats: Compared with the blank group,the index of liver,kidney,spleen and thymus were significantly decreased in the model group(P<0.01),except the thymus index of the low dose group(P>0.05),the index of other groups increased significantly(P<0.05 or P<0.01).3.SOD activity in serum,spleen and kidney: Compared with the blank group,the SOD activity in serum,spleen and kidney of the model group decreased significantly,with a significant difference(P<0.01).Compared with the model group,except the low dose group of spleen(P>0.05),the activity of SOD increased in other groups,showing a significant difference(P<0.05 or P<0.01).4.GSH-Px activity in serum,spleen and kidney: Compared with the blank group,the GSH-Px activity in serum,spleen and kidney of the model group had an extremely obvious decreasing trend,showing a very significant difference(P<0.01).Compared with the model group,the GSH-Px activity of each group increased significantly(P< 0.05 or P<0.01).5.Content of MDA in serum,spleen and kidney: Compared with the blank group,the content of MDA in serum,spleen and kidney in the model group increased significantly,showing a very significant difference(P<0.01).Compared with the model group,the content of MDA in each group decreased significantly(P< 0.05 or P<0.01).6.Histomorphology of kidney in rats: In the blank group,the size and shape of kidney tissue were normal,and there were no obvious pathological changes.In the model group,the distribution of glomerulus was loose and sparse,and the spherules were atrophied and irregular around the periphery.The lumen of glomerulus and renal tubule were significantly expanded.Compared with the model group,the pathological state of vitamin E and Tianxingjianshenwei granule groups with different doses was significantly relieved.Conclusion:1.Tianxingjianshenwei granule can improve the general state of aging rats and slow down the pathological process of organ aging.2.Tianxingjianshenwei granule can increase the activity of SOD,GSH-Px in serum,spleen and kidney,decrease the content of MDA and enhance the ability of antioxidation,delay the aging process.
