| Objective:By observing the effect of Tianxingjianyikang Granule on Antioxidation in aging model rats induced by D-GALactose,in order to provide scientific basis for the anti-aging effect of Tianxingjianyikang granule.Methods:Sixty male SD rats were randomly divided into 6 groups according to the body mass level,with 10 rats in each group:blank group,model group,vitamin E positive control group and low,medium and high dose groups of Tianxingjianyikang granule.The aging model was established by intraperitoneal injection of D-GAL.except the blank group,the other five groups of rats were intraperitoneally injected with D-GAL solution according to the standard of 0.5 g/kg for 42 days.From the 15th day of modeling,the rats in the vitamin E positive control group were given vitamin E solution by gavage according to the standard of 27mg/kg,the low,medium and high dose groups of Tianxingjianyikang granules were given 1.3 5 g/kg,2.7g/kg and 5.4g/kg respectively,and the blank group and model group were given physiologic saline with equal volume ratio once a day for 28 days.During the experiment,the changes of general state of rats in each group were observed;After the experiment,the body mass of 60 rats was weighed for the last time,and the blood was taken from the abdominal aorta after anesthesia;The tissues of brain,liver,spleen and thymus of rats were quickly and completely removed from the ice box,their mass was weighed,and the organ index was calculated;The activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)in serum,liver and brain were measured,and the effects of each group on antioxidant capacity were compared;HE staining sections of rat brain tissue were made to observe the morphological changes of hippocampus.Results:1.Observation results of general state of rats in each group:compared with the blank group,the general state of rats in the model group is poor,with rough,dry and loose skin,lack of luster and unsmooth hair,accompanied by a small amount of falling off,decreasing amount of exercise,correspondingly reducing amount of food and water intake,depressed mental state and so on;Compared with the model group,the general state of rats in other administration groups has been improved to varying degrees,and the state of rats in the control group and high-dose group is the best.2.Measurement results of organ index of rats in each group:compared with the blank group,the organ index of rats in the model group decreased significantly(P<0.01);Compared with the model group,the organ index of rats in each treatment group increased in varying degrees(P<0.01 or P<0.05).The organ index of rats in vitamin E group and Tianxingjianyikang granule high-dose group increased most significantly,which significantly improved the state of organ atrophy.3.Detection results of antioxidant indexes:compared with the blank group,the activities of SOD and GSH-Px in serum,liver and brain of model group were significantly decreased(P<0.01),and the content of MDA was significantly increased(P<0.0 1);Compared with the model group,Tianxingjianyikang granule could significantly increase the activity levels of SOD and GSH-Px in serum,liver and brain tissue of rats in each group(P<0.01 or P<0.0 5),and significantly reduce the content of MDA(P<0.01 or P<0.05).Among them,Tianxingjianyikang granule high-dose group and vitamin E group had the most prominent effect.4.Morphological observation of hippocampal tissue:In the blank group,the hippocampal tissue structure was compact,the morphology of neurons was regular,the staining of cytoplasm and nucleus was clear,there was no congestion or edema and no inflammatory cell proliferation;In the model group,the hippocampal tissue structure was loose,the arrangement of neurons was irregular,the nucleus showed obvious pyknosis,the number of apoptotic bodies,microglia and inflammatory cells increased significantly,and the phenomenon of capillary dilation appeared;Compared with the model group,the histomorphology of hippocampus in each administration group was improved in varying degrees,especially in vitamin E group and Tianxingjianyikang granule high-dose group.Conclusion:1.Tianxingjianyikang granule can improve the general state and organ atrophy of aging rats to a certain extent.2.Tianxingjianyikang granule can increase the activities of SOD and GSH-Px in serum,liver and brain of aging rats,reduce the content of MDA and improve the antioxidant capacity of the body.At the same time,it can improve the tissue morphology of hippocampus of aging rats to a certain extent and has a certain anti-aging effect. |
