Objective:By searching the clinical research literature of Chinese medicine for the treatment of Diabetic kidney disease(DKD)with Qi-Yin deficiency and stasis from the Chinese journal full-text databases(China Knowledge Network,Vipshop,Wanfang)from the year of establishment to 2022,we used data mining research methods to summarize its medication pattern,and combined with network pharmacology research methods to explore the action targets and related pathways of core drugs for the treatment of DKD with Qi-Yin deficiency and stasis,and to explore its mechanism of action for the treatment of DKD with Qi-Yin deficiency and stasis,so as to provide theoretical basis for further clinical treatment and experiments of this disease.Methods:1.In this study,we searched the clinical research literature of Chinese medicine for the treatment of Diabetic kidney disease(DKD)with qi-yin deficiency and stasis by searching the full-text databases of Chinese journals(China Knowledge Network,Vipshop and Wanfang)from the establishment to2022,formulated the inclusion and exclusion criteria,screened the relevant literature,and established the "Chinese medicine database for the treatment of diabetic diabetes with qi-yin deficiency and stasis"."The database was established.Based on the TCM heritage platform,we analyzed the data and mined the high frequency drugs,their nature and ascriptions,prescriptions and combinations through drug statistics,drug clustering,association rules and other data analysis methods,and finally performed visual analysis and display.2.Using TCMSP database to retrieve the chemical composition of core drugs,predict the protein target information,and establish the active ingredienttarget database of core drugs.Retrieve potential targets related to Diabetic kidney disease in Gene Cards and OMIM databases.Take intersecting targets between drug targets and disease targets and construct protein interaction network using String platform.Cytoscape 3.7.2 was used to construct the "active ingredient-target" network map,and DAVID was used to perform GO functional enrichment analysis and KEGG enrichment analysis.Results:1.The analysis of the TCM heritage platform yielded the results of the medication pattern of TCM for the treatment of Diabetic kidney disease with Qi and Yin deficiency and stasis.The results found that the top 5 high-frequency words of TCM were Astragalus,Salvia,Yam,Cornus,and Radix et Rhizoma.Taste was mainly warm,cold,flat,sweet and bitter,and the normalizing meridians were liver,kidney and spleen.Most of them are used to nourish deficiency and invigorate blood circulation and remove blood stasis.With the association rules,those with higher support and confidence were Astragalus,Salvia,Yam,Cornus,Radix et Rhizoma and Poria.Cluster analysis can classify the drugs into 4 categories,1: astragalus,yam,raw dioscorea,danshen,maidenhair and cornelian;2: astragalus,cornelian,yam,zedoary,poria and gorgonian;3: astragalus,danshen,huangjing,yam,raw dioscorea and codonopsis;4: astragalus,danshen,cornelian,yam,princely ginseng and poria.2.Through the network pharmacology method,using the TCMSP database screening,we obtained a total of 75 active ingredients of drugs in the core formula of Sanhuang Yi Kidney Capsule(20 Astragalus,12 Radix et Rhizoma,8 Radix et Rhizoma,15 Yam and 20 Cornu Cervi Pantotrichum),431 action targets,227 genes of intersection between drugs and DKD diseases,and through the drug-active ingredient-target network,we inferred that the core active ingredients of Sanhuang Yi Kidney Capsule KEGG enrichment analysis showed that the core formula of Sanhuang Yi Kidney Capsules for DKD may be related to Estrogen signaling pathway,VEGF signaling pathway,AGE-RAGE,and the Gene Net.pathway,AGE-RAGE signaling pathway in diabetic complications,prolactin signaling pathway,relaxin signaling pathway,and Relaxin signaling pathway,TNF signaling pathway,and Thyroid hormone signaling pathway are closely related.Conclusions:1.The drug use pattern of Chinese medicine in the treatment of Diabetic kidney disease with deficiency of both qi and yin is mainly warm,cold and flat,and the taste of the drugs is mainly sweet and bitter,and the normalization is related to all five organs.The drug categories and properties obtained from this analysis are in accordance with the traditional Chinese medicine theory and clinical practice on the understanding of the etiology and pathogenesis of DKD with Qi-Yin deficiency and blood stasis,and they are consistent with the core drug composition of the clinical basis of the self-prepared formula San Huang Yi Kidney Capsules,which was studied by the group in the previous phase.The composition of the capsule coincides with that of the core drug of the clinical basis of the self-prepared formula San Huang Yi Kidney Capsules,which has the value of further research.2.Using the network pharmacology method and combined with molecular docking technology,the key targets of the core formula of Sanhuang Yi Kidney Capsule for DKD were verified,and it was found that the effects of the core formula of Sanhuang Yi Kidney Capsule for DKD mainly involved AKT1,TNF,IL-6 and other targets,VEGF,AGE-RAGE and other signaling pathways,suggesting that the mechanism of the core formula of Sanhuang Yi Kidney Capsule for DKD may be related to anti-inflammatory,anti-inflammatory,antiinflammatory,anti-drug,anti-drug,anti-drug,anti-drug,anti-drug,anti-drug and anti-drug.mechanism may be related to anti-inflammation,alleviation of oxidative stress and improvement of vascular endothelial injury.
