| Purpose:Through the data mining of the clinical medication literature of the method of nourishing qi,nourishing yin and activating blood for the treatment of diabetic nephropathy,flexibly using the "Traditional Chinese Medicine Inheritance Auxiliary Platform(V2.5)" software,inputting relevant information,and obtaining the medication rules;and then using network pharmacology to study,to explore the effective targets,signaling pathways and mechanisms of action of the core prescriptions in the treatment of diabetic nephropathy,and to provide a good reference standard for clinical precision medicine and animal experiments.Material and method:1 Relevant clinical literatures from January 31,2011 to January 31,2021 were retrieved from CNKI,Wanfang,and VIP databases using computers flexibly.Strictly according to the inclusion and exclusion criteria,the relevant literatures on the treatment of diabetic nephropathy with traditional Chinese medicine for nourishing qi,nourishing yin and activating blood were screened out,and the original Excel2019 original literature database was established;then the "Traditional Chinese Medicine Inheritance Auxiliary Platform(V2.5)" was used for data analysis to obtain drugs An Excel database was established with information such as frequency,four qi and five flavors,core drug combinations,and prescription rules.2 Obtain the active ingredients and corresponding effective target genes of Astragalus,Rehmannia glutinosa and Salvia miltiorrhiza by searching the TCMSP database(for the convenience of description,the three are collectively referred to as traditional Chinese medicine compounds),and apply the disease targets obtained through the Dis Ge NET and Gene Cards databases to obtain DN using Excel.Take the union again,draw the Venn diagram of drug-disease action targets through Venny2.1,and use Cytoscape3.9.0 to make a visual network diagram of traditional Chinese medicine compound-core target-diabetic nephropathy;use String to make PPI network diagram;through DAVID database GO function and KEGG pathway enrichment analysis was performed on the core targets,and the GO and KEGG analysis results were imported into the We Chat website for visual display of bubble charts.Results:data mining results1 Strictly according to the set inclusion and exclusion criteria,the author obtained 201 valid literatures,including 201 first prescriptions,including 162 traditional Chinese medicines.There are 31 traditional Chinese medicines that are used 25 times or more.Astragalus 188 times,followed by Salvia miltiorrhiza and Rehmannia glutinosa.2.According to the statistics of nature and flavor returning to the meridian,the four qi middle-warm medicines are up to 770 times,accounting for 34.34% of the total,followed by cold and flat medicines;the five-flavor middle-sweet taste is the most frequent,reaching 1478 times,accounting for 43.02 of the total.%.Bitterness and pungency are the second;the top three meridians are liver,kidney,and spleen meridians,and the most frequent is liver meridian 1160,accounting for 19.82% of the total.3 Through the analysis of association rules,the number of support degrees was adjusted to"50",and the confidence level was adjusted to "0.9",and 36 drug combinations with a frequency of 50 and above were obtained,and the top three core drug pairs were "Rehmannia glutinosa".-Astragalus" 121 times,"Salvia-Astragalus" 120 times,"Astragalus-Chinese yam" 106 times,the highest confidence is "Shenyu,Rehmannia glutinosa-> Astragalus" and"Rehmannia glutinosa,Salvia,Chinese yam-> Astragalus" are both 1.4 According to the experience of predecessors and the combination of their own,set the correlation degree to "8",the penalty degree to "2",and the Result value to 2.Through the combination of drugs with high correlation degree,cluster analysis can be used to obtain potential Muxiang,Asparagus,Aconite;Muxiang,Aconite,Cynomorium;Shiwei,Sangjiji,Eucommia;Ophiopogon japonicus,Zelan,Alisma,etc.16 groups of core formulas,on this basis,are related to each other to obtain Maimuxiang,Asparagus,Aconite,Cynomorium;Shiwei,Sangji,Eucommia,Dendrobium;Ophiopogon japonicus,Zelan,Alisma,Cortex Moutan;Rehmannia glutinosa,dogwood,yam,licorice and other 9 groups of new prescriptions.Network Pharmacology Results1 The active components of Astragalus,Rehmannia glutinosa,and Salvia miltiorrhiza were obtained through the TCMSP database,respectively 20,2,and 65,a total of 87 active components;the corresponding targets were 195,23,and 137.After strict screening to remove duplicate values,the final target was 234 The target gene for diabetic nephropathy is3737,and 157 core targets were finally obtained through venny2.1 in "Chinese Medicine Compound-Diabetic Nephropathy".2 According to Cytoscape 3.9.0 software,77 effective core components of"Astragalus-Rehmannia glutinosa-Salvia" were obtained,including quercetin 108,kaempferol50,luteolin 49,tanshinone IIA31,coniferin 11,campesterol 2 and so on.plays a pivotal role in the treatment of DKD.Through the STRING database,the PPI network graph was constructed,and 157(nodes,"709 edges were obtained;the results were imported into Cytoscape 3.9.0 to obtain the key targets of traditional Chinese medicine compounds such as JUN,RELA,STAT3,ESR1,etc.3 The DAVID database was used to carry out GO function enrichment analysis on the key target genes of "TCM-diabetic nephropathy",and a total of 240 biological processes were obtained,involving cytokine-mediated pathways,positive regulation of gene expression,and RNA polymerase II promoter transcription.Positive regulation,etc.;the total number of cellular components is 73,including extracellular space,macromolecular complexes,mitochondria,etc.;the total number of molecular functions is 131,involving enzyme binding,protein binding,transcription factor binding,etc.KEGG enrichment showed that the highest pathways involved AGE-RAGE signaling pathway,endocrine resistance,cancer pathway,lipid and atherosclerosis pathway,etc.prescriptions.Conclusion:1 Based on literature data mining,Astragalus,Rehmannia glutinosa,Salvia miltiorrhiza and Chinese yam are the core components of the traditional Chinese medicine compound for nourishing qi,nourishing yin and activating blood in the treatment of DKD patients.2 According to the research of "Astragalus-Shengdihuang-Salvia" in the treatment of DKD,the active ingredients such as quercetin,kaempferol,tanshinone IIA,and campesterol in the drug can act on diabetic nephropathy JUN,ESR1,Target genes such as TNF can regulate the most important signaling pathways such as AGE-RAGE in diabetic nephropathy and achieve the effect of treating DKD.Provides new thinking and animal experiments for our clinical precision medicine integration of traditional Chinese and Western medicine.From this perspective,it has laid a solid foundation for Chinese medicine to go global. |
