| Objective To observe the effect of Wuhe xuduan on the prevention and treatment of scopolamine(SCOP)induced Alzheimer’s disease(AD)model rats with cognitive memory impairment and learning ability,and to explore its possible mechanism of action.Method Rats were divided into a 2-week group and a 4-week group,with each group further divided into a normal group,SCOP group,Western medicine group,and low,medium,and high dose groups of Wuhe xuduan.The two week and four week groups of Wuhe xuduan treatment group were treated with different concentrations of water decoction according to the treatment time;The blank control group and SCOP group were administered the same volume of warm double distilled water.The rats in the 2-week group received water maze training on the 9th day of gavage for 5 consecutive days.The water maze rested on the 14 th day and was injected intraperitoneally at 10 am on the 15 th day to create a model.The model was created using intraperitoneal injection 0.5 hours before the training,followed by the first quadrant water maze test.The survival platform was removed during the testing experiment and the number of times the rats crossed the platform quadrant was recorded;All groups of rats in the 4-week group underwent water maze training on the 23 rd day after gavage.The rats were gavaged 0.5 hours before each training session,and a model was created by intraperitoneal injection 0.5 hours before the training session.The water maze was continuously trained for 5 days,rested on the28 th day,and injected intraperitoneally at 10 am on the 29 th day.The first quadrant water maze test was conducted every 0.5 hours after the model was established;After the test,the tissues were taken for Nissl staining to detect the number of Nissl bodies,and Golgi staining to detect the number and morphology of dendritic spine;Cut off the head and take the hippocampus for acetylcholine transferase content detection,protein immunoblotting technology to detect Tau protein and synaptic related protein levels in the hippocampus.Result In both the 2-week and 4-week groups of the treatment group,compared to the model group,the Western medicine group and the high-dose group of Wuhe xuduan significantly shortened the time to escape the incubation period and increased the number of crossing platforms,proving that the Western medicine and high-dose group of Wuhe xuduan have better therapeutic effects;Immunoblotting showed that after administration of Wuhe xuduan,the Western medicine group and high-dose Wuhe xuduan group in the 2-week and 4-week groups showed better therapeutic effects,reducing the phosphorylation level of Tau protein and increasing the levels of related synaptic proteins in the hippocampus:Synaptophysin,P-Synapsin(Ser9),PSD95,P-Ca MK II α(Thr286);The number of dendritic spine and Nissl bodies could be increased in the western medicine group and the high dose group of Wuhe xuduan in the 2-week and 4-week groups.Conclusion Wuhe xuduan can improve cognitive memory impairment and learning ability in SCOP induced AD model rats,and has a preventive and therapeutic effect on AD.The possible mechanism is to increase the activity of acetylcholine transferase,regulate the cholinergic system,reduce the hyperphosphorylation of Tau protein,increase the expression level of synapse related proteins,promote the improvement of the number and morphology of dendritic spine,increase the number of Nissl bodies,protect neurons,and achieve the effect of prevention and treatment of AD. |
