[Aim] To investigate whether Radix astragali induces changes and preliminary mechanisms of action in learning and memory of model rats with Alzheimer disease.[Methods] Totally 40 Wistar rats were inclued in the experiment and randomized into 4 groups, negative group, masculine group (piracetam), Radix astragali low dose group and Radix astragali large dose group,10 rats in each group. The model rats with Alzheimer disease were made by injecting Aβ1-40 (4μg) in hippocampus after intraperitoneal injecting D-galactose (50mg/kg.d) six weeks. The learing and memory were detected by Morris water maze after intragastric administration two weeks. The concentration of acetylcholine esterase was determined via the method of biochemistry.[Results]1. Effects of Radix astragali on common condition and weight in rats:After intragastric administration two weeks with Radix astragali in rats, the common contion were good. Moreover, all Radix astragali groups's rats weight hadn't significantly changed compaired with negative group (P>0.05).2. Effects of Radix astragali on learning and memory:①Radix astragali with low dose and large dose can shorten ascend platform latent period with negative group in rats (P<0.05).②Radix astragali with low dose and large dose can increase crossing platform number of times with negative group in rats (P<0.05).3. Effects of Radix astragali on the concentration of acetylcholine esterase in rat hippocampus:Radix astragali with low dose and large dose can increase the concentration of acetylcholine esterase compaired with negative group (P<0.01).[Conclusion] It is indicated that Radix astragali provides good intervention on learning and memory of rat model of Alzheimer disease via enhancing the concentration of acetylcholine esterase in rat hippocampus.
