Objectives:Obesity has a close relationship with the occurrence and development of metabolic diseases such as insulin resistance,Type 2 Diabetes,hyperlipemia and non-alcoholic fatty liver disease.Obesity and other metabolic diseases have systemic low-grade inflammation,but the pimelitis can induce and maintain the systemic low-grade inflammation,which is an important component.Fecal Microbiota Transplants is applied into many diseases,the application in the obesity and metabolic diseases should be closely watched.For the research on obesity treatment through fecal microbiota transplantation and metabolic diseases,its mechanism of action is not clear.The research,through the research on the influence of fecal microbiota transplantation on intestinal flora of obese rats and T cell-mediated pimelitis,firstly discusses action and mechanism and provides experiment and theoretical basis for obesity treatment through fecal microbiota transplantation and diseases related to the obesity.Methods:26 male SD rats(weight 99.2 ± 6.9 g)were randomly divided into normal control group(NG,n=6)and obesity group(n=20).The normal control group was fed with maintenance feed.The obesity group was fed with high fat diet for 8 weeks.10 obese rats whose body weight was20% higher than that in the normal control group were randomly divided into two groups,namely obesity group(HGA,n=5)and obesity group with fecal microbiota transplantation(HGB,n=5).The fecal microbiota in NG group were transplanted into HGB group,and NG group and HGA group were given normal saline.After treatment,body mass,body length and Lee’s index were measured,intestinal flora were detected by 16 S r DNA sequencing,plasma diamine oxidase(DAO)level was detected by double antibody sandwich method,plasma LPS content was detected by ELISA,and Th17 and Treg expression in adipose tissue were detected by immunohistochemistry.Results:After high fat diet for 8 weeks,the obesity rat model was success.16 S r DNA was used to check rat intestinal flora.It was found that composition of intestinal bacteria for obese rats changed significantly,biodiversity reduced,content of plasma DAO and LPS increased,adipose tissue Th17 expression increased,and Treg expression reduced.After fecal microbiota transplantation,weight of obese rats significantly reduced,intestinal flora abundance changed,and biodiversity returned to some extent.The content of Plasma DAO and LPS reduced,Th17 expression of the adipose tissue reduced,and Treg expression increased.Conclusions:1.The intestinal flora of obese rats changed,and obesity had a correlation with the intestinal flora.The adipose tissue had T cellular infiltration and there was pimelitis.2.fecal microbiota transplantation can return intestinal flora of obese rats,increase intestinal mucosal barrier,reduce intestinal permeability,decrease endotoxemia of obese rats,reduce Th17 expression,increase Treg expression of adipose tissue for obese rats,adjust Th17/Treg balance,and suppress inflammation of adipose tissue.
