| Objective: To explore the prescription rule of heat-clearing and blood-cooling method in the treatment of immune thrombocytopenia(immunethrombocytopenia,ITP)by using SPSS software,and to explore its action mechanism of core drugs with network pharmacology,so as to provide useful reference for the treatment of modern blood-heat syndrome ITP disease,in order to provide reference for clinical practice.Methods:1.Data mining: take the Chinese Medical Code as the collection object,search the related TCM disease names of ITP as keywords,collect the prescriptions with the method of clearing heat and cooling blood as the main treatment and establish the database;input the data into SPSS for frequency statistics,cluster analysis,association rule analysis,and get the core drug combination.2.Network pharmacology: through the Chinese Herbal Medicine system Pharmacology platform(TCMSP)database to screen the chemical components of the core drugs and predict the potential action targets,screen the ITP-related targets in the genetic database,and construct the drug composition-ITP-target network.The drug-disease PPI network was constructed by String,and the core targets were screened.The results obtained from the mapping between the active components of the core traditional Chinese medicine and the target of immune thrombocytopenia were introduced into the online software David for GO and KEGG analysis to explore the signal pathway of the core drug components in the treatment of ITP and to construct the drug component-key target-pathway network.Results: 1.Data mining results: 1)High-frequency drugs: a total of92 prescriptions and 190 kinds of drugs for ITP were collected,and the total frequency of drugs was 877.The high-frequency traditional Chinese medicines used include Scutellaria baicalensis,Gardenia jasminoides,Rhinoceros horn,Radix Rehmanniae,Radix Scrophulariae,Cimicifugae,Gypsum,and Rhizoma Coptidis.2)three groups of clustering prescriptions were obtained by cluster analysis: C1: Radix Rehmanniae,Peony bark,Rhinoceros horn,Forsythia suspensa,Honeysuckle,Scrophulariae,Red peony,Coptis chinensis,Lophatherum gracile brongn,gypsum,Anemarrhena asphodeloides,ginseng.C2:Clearing heat and relieving external symptoms: Sophora flavescens,mint,burdock seed,Angelica sinensis,white peony,Fangfeng,Jingmustard.C3:Qingre Xiehuo category: Glauber’s salt,rhubarb,cohosh,Folium Isatidis,Scutellaria baicalensis,Gardenia jasminoides,Semen Sojae Praeparatum.3)by analyzing the association rules of traditional Chinese medicine,it is found that the common medicine pair is Rehmannia glutinosa + Cortex moutan,and the common combination of three drugs is Radix Rehmanniae + Cortex moutan + rhinoceros horn,and the degree of support is 16.30% and 14.13% respectively.4)combined with frequency,association rules,cluster analysis and famous experience,the core drug combination is obtained: Radix Rehmanniae + Cortex moutan +rhinoceros horn.2.Network pharmacological results: 1)A total of 60 active components of core drugs,including quercetin,paeoniflorin,betulinic acid,etc.,were obtained from Disgenet database and Gene Cards database,respectively.4537 targets related to immune thrombocytopenia were obtained after removing duplicates.The gene data set screened from ITP targets and core traditional Chinese medicine targets were introduced into Wayne map.2)Visualization of the relationship between the active components of traditional Chinese medicine and the target,the drug ingredients with the highest D value are quercetin,kaempferol,Radix Rehmanniae violoside A,paeoniflorin and so on.3)construct PPI network and obtain key targets such as AKT1,VEGFA,STAT3 and so on.4)the results of GO analysis showed that there were 887 pathways and 156 pathways in KEGG,suggesting that the pathways involved in the treatment of ITP with core drugs include PI3K-Akt signal pathway,tumor-related micro RNAs signal pathway,TCR signal pathway and so on.Conclusions: 1.Based on data mining "Chinese Medical Code",the core drug combination of clearing away heat and cooling blood in the treatment of ITP is Radix Rehmanniae + Cortex moutan + rhinoceros horn.2.The core drug combination treats ITP through complex signal pathways with multiple components and multiple targets.The main mechanism may be related to regulating immunity,reducing platelet destruction and promoting the growth of vascular endothelial cells. |
