Objective:Statistical analysis of the dosing pattern and network pharmacology of Chinese herbal medicine for the treatment of large intestine damp-heat type UC using data mining methods to explore the mechanism of action of core drugs for the treatment of UC.Methods:(1)We searched the clinical research literature on Chinese herbal medicine for the treatment of large intestine damp-heat type UC published in Chinese Biomedical Literature Network,Wanfang,China Knowledge Network,and Vipro Pubmed Chinese and English databases since the establishment of the database to December 2022,and established the Chinese medicine database,and analyzed the drugs by frequency,flavor attribution,and clustering by the TCM heritage computing platform software,and used SPSS Modeler 18.0 software The association rule analysis was performed.(2)Using network pharmacology methods to analyze the active compounds of core drug combinations,predict potential targets,collect disease targets of UC,obtain drug-disease common targets,and perform GO functional enrichment analysis and KEGG pathway enrichment analysis on drug-disease common targets to analyze the mechanism of action of core drug combinations for the treatment of UC from the perspective of network pharmacology.Results:A total of 150 papers meeting the criteria,159 prescriptions and 193 herbal medicines were included in this study.(1)Combined with the analysis of the overall prescription study,the 10 herbs with the highest frequency of use were Baitou wang,Gan Cao,Mu Xiang,Huang Qin,Huang bo,Di Yu,Bai He,Bai Shao,and Qin Pi.(2)Among the various types of Chinese medicine,the most frequently used type of Chinese medicine is heat-clearing medicine,and the rest are,in order,deficiency tonic,hemostatic,qi-regulating,and water-releasing and dampness-permeating.(3)In terms of nature flavor and meridian: cold drugs were used most frequently 926 times,accounting for 54.2% of the frequency ratio;the rest were warm drugs(395,23.1%),flat drugs(209,12.2%),cool drugs(54,3.2%),and hot drugs(23,1.3%)in that order.In terms of meridians,the first ranking was liver and stomach meridians,and the rest were spleen,large intestine and lung meridians in that order.(4)Cluster analysis: 3 core drug combinations were derived for cluster analysis,of which the largest number of clustered prescriptions was the combination: Baitouwong,Huanglian,Huangbo,Qinpi and Gancao,and the number of clustered prescriptions obtained was 68,accounting for 42.80%.(5)Association rule results: The 193 Chinese herbal medicines studied were analyzed by association rules,setting the parameter threshold: "The minimum number of support is 10%,and the confidence level was set to 80%.34 association rules between drugs were obtained,among which the first ranked combinations of two,three,and four drug pairs were: Baitouweng → Qinpi,Qinpi + Huangqin → Baitouweng,Qinpi + Huangbo + Huangqin → Baitouweng.(6)The core drug combination in the overall prescription,"Baitou weng,Huanglian,Huangbo,Huangqin,Qinpi and Gancao"(i.e.Baitou weng plus Gancao soup),was collected through the network pharmacology method with 123 active ingredients,70 potential targets,31 drug-disease common targets,458 GO-BP,33 GO-MF,21 GO-CC after GO functional analysis,66 KEGG pathway enrichment analysis and 9 molecular docking of major active compounds and major targets.Conclusion(s):In this study,the data mining method was used to reveal that traditional Chinese medicine treatment of large intestine damp-heat type of UC is mainly based on cold,bitter,diarrhea to clear heat and dry dampness,supplemented by warming and calming to regulate qi and tonify deficiency,while attributing to liver,stomach,spleen,large intestine and lung meridians as the key,in order to highlight the drug use pattern of clearing heat and dampness,detoxifying and stopping dysentery,moving qi and cooling blood,and strengthening the spleen and stomach,and investigated the potential mechanism of action of the core drug formula Baitou Weng plus Gancao Tang for the treatment of UC,which may be through The potential mechanism of action of the core drug formula Baitouweng plus Gancao Tang in the treatment of UC may be through the action of the active ingredients such as β-sitosterol,quercetin,flavopiridol,chrysin and isorhamnetin on the core target genes such as TP53,CASP3,CASP8,CASP9,CAT,CCL2,BCL2L11,CAV-1,MMP2,NR3C1,which regulate the biological processes such as inflammatory response and oxidative stress,and further regulate TP53,apoptosis and other The effect of the signaling pathway,such as anti-inflammatory and analgesic,antibacterial,anti-oxidative stress,anti-tumor,protection of intestinal mucosa,etc.,so as to achieve the therapeutic effect of the treatment of UC. |
