| Objective:To explore the clinical efficacy of the method of nourishing Qi and Yin in treating the syndrome of Qi and Yin deficiency in chronic stable exertional angina pectoris,and its antagonistic effect on endothelial progenitor cell injury,so as to reveal the protective mechanism of the method of nourishing Qi and Yin on vascular endothelium in chronic stable exertional angina pectoris.To provide the basis for the clinical application of traditional Chinese medicine.Methods:1.Clinical study:Forty patients meeting the criteria were screened,which were divided into control group and treatment group randomly.The control group was treated with basic treatment and Nuoxinkang,and the treatment group was additionally treated with Yiqi Fumai injection on the basis of this.The treatment period was 10days.The improvement of angina pectoris score,TCM syndrome score,electrocardiogram effect and safety index were observed before and after treatment,and the clinical efficacy and safety were evaluated.2.Clinical basic research:Forty patients meeting the criteria were screened,that had been treated with conventional treatment and were given an additional Yiqi Fumai injection.Meanwhile,20 healthy volunteers were included as the normal group,and no intervention was performed on them.Fasting venous blood was collected from each subject in the morning on the day of inclusion and 10 days after intervention.The number of endothelial progenitor cells in peripheral blood was counted by flow cytometry,and the phenotypic markers were CD34~+CD133~+and CD34~+KDR~+.Peripheral blood endothelial progenitor cells were cultured in vitro,identified by fluorescence staining,and cell adhesion,migration,tubule formation,and proliferation were tested.Results:1.Clinical study:the treatment group was better than the control group in improving the efficacy of TCM syndromes,reducing the scores of TCM syndromes and the score of angina(P<0.05).The treatment group was similar with the control group in terms of angina and electrocardiogram efficacy improvement(P>0.05).There were no abnormal changes in the safety index before and after treatment(P>0.05).2.Clinical basic research:the number of endothelial progenitor cells in peripheral blood of patients with chronic stable exertional angina pectoris was significantly lower than that of healthy volunteers(P<0.05).After 10 days of treatment,the number of endothelial progenitor cells in peripheral blood of patients with chronic stable exertional angina pectoris was significantly higher than before treatment(P<0.05).The migration,adhesion,tubular formation and proliferation of cultured peripheral blood endothelial progenitor cells in patients with chronic stable exertional angina pectoris were impaired,and their functions were significantly improved after treatment with Yiqi Fumai injection.Conclusion:the method of nourishing Qi and Yin can improve the efficacy of TCM syndrome in patients with chronic stable labor angina pectoris with deficiency of both Qi and Yin,and reduce TCM syndrome and angina score.The mechanism is related to its antagonism against endothelial progenitor cell injury,thus improving vascular injury and endothelial function. |
