Objective To study the basic characteristics,similarities and differences of airway microecology between patients with acute exacerbation of chronic obstructive pulmonary disease(AECOPD)and healthy people in Inner Mongolia,and to analyze the correlation between airway microecological characteristics and clinical indicators of AECOPD patients.The research on airway microecology provides relevant data and expands new ideas for the diagnosis and treatment of diseases.Methods Questionnaire survey was used to record general information and make statistics on relevant clinical indicators.Sputum samples were collected from 36 healthy volunteers and 34 patients with AECOPD,and 16 S r DNA high-throughput sequencing was used to analyze the airway microecological structure.The airway microecological characteristics and differences between healthy people and AECOPD patients were revealed from the aspects of alpha diversity,beta diversity and Lef Se difference analysis,and the correlation between airway microecological characteristics and clinical indicators of AECOPD patients was analyzed.Results 1.There are differences in the airway microecological structure between healthy people and AECOPD patients in Inner Mongolia.The alpha diversity of airway microecology in healthy population was higher than that in AECOPD population.Beta diversity analysis:PC1 contribution rate was 50.31% and PC2 contribution rate was 14.91% based on the principal coordinate(PCo A)analysis of Weighted_Uni Frac distance.Lef Se analysis of the airway differences between healthy people and AECOPD showed that at the phylum level,the relative abundance of Bacteroidetes in the healthy group increased;at the genus level,Pseudomonas,Veillonella(Veillonella),Burkholderia(Burkholderia)relative abundance increased compared with healthy group,while Neisseria(Neisseria),Prevotella(Prevotella),Haemophilus(Haemophilus),Pseudobacterium The relative abundances of Alloprevotella and Porphyromonas were decreased compared to the healthy group.2.Long-term use of inhaled glucocorticoid + long-acting β2-receptor agonist mixture(ICS + LABA)in the AECOPD population will reduce alpha diversity.Procalcitonin(PCT),blood mononuclear cell count(MONO),hemoglobin(HGB),D-dimer(D-D)and body temperature were negatively correlated with the alpha diversity of airway microecology,and the difference was statistically significant Significance(P<0.05).Conclusion The airway microecological composition of the AECOPD population in Inner Mongolia was different from that of the healthy population,and the airway microecological diversity decreased.Long-term use of ICS+LABA formulations in the AECOPD population decreased alpha diversity.Alpha diversity in AECOPD patients was negatively correlated with inflammatory markers(PCT,MONO,D-dimer,body temperature)and HGB.This experimental study provides microecological data support for the airway microecology of healthy and AECOPD populations in Inner Mongolia,and also provides new ideas for the diagnosis and treatment of AECOPD.
