Objective The purpose of this study is to explore the level of nuclear factor Interleukin 3(NFIL3)m RNA in colorectal cancer through bioinformatics methods combined with experiments,to analyze the relationship between NFIL3 level and the clinical pathological characteristics and prognosis of colorectal cancer,to explore the clinical significance of NFIL3 in colorectal cancer,and to provide a basis for the selection of molecular targets for the detection of molecular markers for colorectal cancer and the determination of prognosis.It provides new ideas for the future study of the molecular mechanism of colorectal cancer.Methods 1.Based on TCGA database,download colorectal cancer NFIL3 m RNA data in TCGAs,clinical pathological characteristics data of colorectal cancer patients(including: sex,age,degree of tumor invasion,lymph node metastasis,distant metastasis,clinical stage)and overall survival(OS).The level of NFIL3 m RNA in colorectal cancer tissues,the relationship between NFIL3 expression and the clinicopathological features of colorectal cancer and the correlation with the prognosis of colorectal cancer were analyzed.2.Fresh specimens of cancer tissue and paracogenic normal tissue(> 5 cm from the edge of the mass)were continuously collected from March 2021 to May 2021 in the Department of Anorectal Disease of Chifeng Clinical Medical College of Inner Mongolia Medical University for radical surgery of colon(rectal)cancer.Quantitative Real-time PCR(q RT-PCR)was used to detect the relative levels of NFIL3 gene m RNAs in collected specimens,and the correlation between NFIL3 m RNA levels and patients’ clinical pathological characteristics(including sex,age,tumor diameter,TNM stage,clinical stage),preoperative laboratory indicators(CEA,CA-199,NLR)and KRAS-E2 mutation status were analyzed.Results 1.Download the colon cancer and rectal cancer related data in the TCGA database,including 698 samples,including 480 colon cancer samples,167 rectal cancer samples,and 51 normal/paracancerous samples for analysis,and the results showed that the level of NFIL3 m RNA was significantly different in colorectal cancer and normal or paracancerous tissue,and lower in colorectal cancer tissue(P<0.05).2.The analysis of NFIL3 levels and clinicopathological features in colorectal cancer samples showed that there were significant statistical differences between NFIL3 m RNA levels and tumor patients’ age of onset,pathological T stage and clinical stage(P<0.05),and there were no statistical differences between NFIL3 expression and tumor patient sex,lymph node metastasis and distant metastasis in color cancer samples(P>0.05).3.The results of survival analysis showed that the level of NFIL3 m RNA was significantly correlated with OS in colorectal cancer patients,and the OS in colorectal cancer patients with high NFIL3 m RNA level was significantly shortened(P<0.05),NFIL3 was analyzed by Cox regression model to be an independent risk factor for colorectal cancer prognosis.4.The results of q RT-PCR in clinical specimens suggested that the level of NFIL3 gene m RNA in colorectal cancer tissue was significantly lower than that of the corresponding paracancerous normal tissue(P<0.05);the NFIL3 m RNA level of cancer tissue,clinical characteristics and laboratory indicators were statistically analyzed,and the results showed NFIL3 High levels of m RNA were significantly associated with T stage of tumor pathology and clinical stage in colorectal cancer(P<0.05)and with CEA(P<0.05),but not with patient sex,age,tumor diameter,lymph node metastasis,distant metastasis,CA19-9,NLR value,and KRAS-E2 mutation status.Conclusion 1.The level of NFIL3 m RNA is significantly reduced in colorectal cancer tissues,and NFIL3 may be significantly related to the occurrence and development of colorectal cancer.2.The level of NFIL3 m RNA in colorectal cancer tissues was significantly correlated with the age of onset,TNM stage,clinical stage and CEA level of colorectal cancer patients,and NFIL3 was involved in the invasion and metastasis of colorectal cancer,providing an option for detecting biological markers of colorectal cancer.3.The OS of colorectal cancer patients in the NFIL3high-expression group was significantly shortened,which was an independent risk factor for poor prognosis in colorectal cancer patients.
