| Objective: To investigate the expression of sterol oxygen-acyltransferase 1(SOAT1)in hepatocellular carcinoma and its relationship with clinicopathological features,and to evaluate the application value of SOAT1 in the diagnosis of hepatocellular carcinoma by combining various markers(HSP70,GS,CD10,CD34).Methods: Retrieve The Cancer Genome Atlas(TCGA)in patients with hepatocellular carcinoma related data,analyze SOAT1 gene in hepatocellular carcinoma tissues m RNA level,and its relationship with HCC clinical characteristics,enrichment,build and use the gene protein regulation network preliminarily SOAT1 genes involved in gene regulation and cancer related signaling pathways.Collect HCC tissue and liver tissue adjacent to carcinoma and benign liver tissue samples,making tissue microarray,respectively from the level of genes(RT-PCR method,RNAscope method)and protein levels(immunohistochemical staining method),analyzes common SOAT1 expression in HCC tissue and its correlation with clinical pathological features.The application value of SOAT1 in the diagnosis of hepatocellular carcinoma was evaluated by immunohistochemical staining combined with multiple markers(HSP70,GS,CD10 and CD34).Results: 1.TCGA database data analysis showed that the m RNA level of SOAT1 gene in hepatocellular carcinoma tissues was significantly higher than that in normal tissues(P<0.01);The m RNA level of SOAT1 gene was correlated with gender of HCC patients(P<0.01);The overall survival rate of high SOAT1 m RNA expression group was lower than that of low SOAT1 m RNA expression group(P<0.01);Univariate and multivariate Cox regression analysis showed that the aged group(P<0.01),male group(P<0.05),and SOAT1 m RNA expression up-regulated group(P<0.05)all suggested poor prognosis.Analysis of gene action pathways based on data sets showed that SOAT1 is involved in gene regulation and cancer-related signaling pathways,and interacts with some proteins to participate in gene monitoring pathways.2.Experimental results: The expression of SOAT1 gene and protein were up-regulated in hepatocellular carcinoma tissues,with significant difference(P<0.01);The lower the differentiation of hepatocellular carcinoma tissues,the higher the protein expression level of SOAT1(P<0.05).The expressions of HSP70,CD10 and CD34 were different in hepatocellular carcinoma tissues and paracancer tissues(P<0.01).The expressions of HSP70 and CD10 in hepatocellular carcinoma and benign liver disease were different(P<0.01).SOAT1 was correlated with the expression of HSP70,GS and CD34(P<0.01).The positive coincidence rate of SOAT1 hepatocellular carcinoma was 63.79%.The coincidence rate of negative diagnosis in paracancer tissue and benign liver disease tissue was 100% respectively.Conclusions: Bioinformatics analysis showed that SOAT1 was involved in gene regulation and cancer-related signaling pathways,providing a research direction for further revealing the mechanism of action of SOAT1 in the occurrence and development of hepatocellular carcinoma.Both gene and protein levels confirmed that the expression level of SOAT1 was up-regulated in HCC and higher in poorly differentiated HCC,suggesting that SOAT1 can be used as a prognostic indicator of HCC.SOAT1,as a marker for benign and malignant hepatocellular proliferative lesions,has good sensitivity and specificity,and its combination with HSP70,GS,CD10 and CD34 has certain application value in the diagnosis of hepatocellular carcinoma. |
