Objective:To investigate the relationship between the expression of SREBP1and SOAT1 and the clinicopathological features and prognosis of hepatocellular carcinoma,and to analyze the correlation between the expression of SREBP1 and SOAT1 in hepatocellular carcinoma.Methods:The data of hepatocellular carcinoma were downloaded from the TCGA(Cancer Genome Atlas)database,and the expression of SREBP1 gene was analyzed by bioinformatics.Collect 96 cases of hepatocellular carcinoma and adjacent liver tissue samples from the clinical pathology department of the First Affiliated Hospital of Hainan Medical College,and make tissue chips.The expression of SREBP1 m RNA and SOAT1m RNA was detected by RNAscope technology.The expression of SREBP1 and SOAT1 was detected by immunohistochemical staining.The relationship between the expression of SREBP1 and SOAT1 and clinicopathological features and prognosis was analyzed.The correlation between the expression of SREBP1 and SOAT1 in hepatocellular carcinoma was analyzed.Results:1、RNAscope detection and statistical analysis results:The expression of SREBP1 m RNA and SOAT1m RNA in hepatocellular carcinoma was significantly higher than that in adjacent liver tissues(P<0.001).There was a positive correlation between the expression of SREBP1m RNA and SOAT1m RNA in hepatocellular carcinoma(P<0.05).2、Immunohistochemical detection and statistical analysis results:The expression of SREBP1 protein and SOAT1 protein in hepatocellular carcinoma was significantly higher than that in adjacent liver tissues(P<0.001);the expression levels of SREBP1 protein and SOAT1 protein were correlated with the differentiation,distant metastasis or local recurrence of hepatocellular carcinoma(P<0.05).The expression of SREBP1 protein was up-regulated in the high HBV-related HCC virus DNA load group(>10~3IU/ml)(P<0.05).The expression of SREBP1 protein was positively correlated with that of SOAT1 protein in hepatocellular carcinoma(P<0.05).Conclusions:1、The expression of SREBP1 and S0AT1 in hepatocellular carcinoma was correlated,and both showed a high expression trend.It provides direction and theoretical basis for the study of lipid metabolism regulation mechanism related to the occurrence and development of hepatocellular carcinoma.2、The expression of SREBP1 protein is correlated with the replication activity of hepatitis B virus,which provides a new research direction for the exploration of the mechanism of the occurrence and development of hepatitis B virus infection-related liver cancer.
