Objective: This study aim to observe the effects of scalp acupoint pressing on neural function and the expression of neurotrophic pathway proteins BDNF,Trk B and CREB in the affected cortex of rats with cerebral ischemia injury and explored the possible mechanism of scalp acupoint pressing on cerebral blood deficiency injury.Methods: SD rats were selected to establish the model of cerebral ischemia injury by longa suture method,and the model was evaluated by Zea longa neurological function score.The 36 successful rats were randomly divided into sham operation group,model group and treatment group,with 12 rats in each group.The treament group was treated by pressing "Baihui" and "shenting",and the time of each treatment is 10 minutes,once a day,for total of 7 days.The other two groups were fed normally.After treatment,the neurological function of rats was evaluated by modified neurological severity scale(MNSs).TTC staining was used to detect the infarct area of rat brain tissue,Nissl staining used to detect the survival of neurons,Western blot used to detect the protein expression levels of BDNF,Trk B and CREB in the ischemic cortex and PCR used to detect the expression levels of BDNF m RNA,Trk B m RNA and CREB m RNA.Results: After intervention,there was significant difference in MNSs score between treatment group and model group(P < 0.05).TTC staining showed that there were obvious pale infarcts in the model group compared with the sham group(P < 0.05);Compared with the model group,the pale infarct area in the treatment group was significantly reduced(P <0.05).Compared with the sham group,the number of nissl bodys in the model group was decreased significantly(P < 0.05);Compared with the model group,the number of nissl bodys in the treatment group was increased significantly(P < 0.05).Western blot showed that compared with the sham group,the expression levels of BDNF,Trk B and CREB proteins in the cortex of the rats in model group was decreased(P < 0.05);Compared with the model group,the expression levels of BDNF,Trk B and CREB proteins in the cortex of the rats in treatment group was increased(P < 0.05).The results of PCR showed that compared with the sham group,the expression levels of BDNF m RNA,Trk B m RNA and CREB m RNA in the cortex of the rats in model group was decreased(P < 0.05);Compared with the model group,the expression levels of BDNFm RNA,TRKBm RNA and CREBm RNA in the cortex of the rats in treatment group was increased(P < 0.05).Conclusion: 1.The intervention of scalp point pressing on rats with cerebral ischemic injury can reduce the damage degree of nerve cells and the infarct state of brain tissue,improve the defect state of nerve function and protect rats from ischemic brain injury.2.Scalp point pressing can regulate the expression of neurotrophin signal pathway BDNF / Trk B / CREB in rats with cerebral ischemia injury.BDNF / Trk B / CREB signal pathway may be the mechanism of scalp point pressing to improve the state of neurological deficit in rats.3.Scalp acupoint tapping based on the theory of "head Qi has Street" and "pressing it leads to hot Qi",which has a protective effect on rats with cerebral ischemia injury. |
