Objective:The experiment will study the learning and memory ability of AD rats and the quantity of BDNF,TrkB and CREB in order to study the therapeutically effect and mechanism of the electroacupuncture(EA) on Alzheimer disease(AD), which will offer experimental basis of applying EA in clinic practice of the acupuncture-moxibustion.Methods:The Alzheimer disease rats were modeled by injectingβ-amyloid protein into bilateral hippocampus of rats and treated by the electroacupuncture therapy of Fengfu.All the experimental rats were divided into four groups:Normal Group, Sham-modeling Group,Model Group,Electroacupuncture Group. (1)Observation on behaviors:Model before the model as well as the end of treatment after the Morris water maze after the experiment,rats escape latency and 60s across the former platform in the frequency of AD in order to understand learning and memory in rats.(2 Histochemical methods used in each group to observe the morphological changes in rats hippocampus.(3) Using immunohistochemical methods hippocampal region of rats in each group brain-derived neurotrophic factor(BDNF) and its receptor tyrosine kinase B(TrkB),cAMP response element binding protein(CREB)-positive cells.Result:(1) In EA group,the number of the learning and memory ability after the treatment was higher than before the treatment(P<0.05),but that didn't change in AD group(P>0.05). (2) the number of BDNF,TrkB and CREB positive neurons in AD group was obviously less than those of the normal group and sham-operation group(P<0.01).In the EA group,the number of BDNF,TrkB and CREB positive neurons were more than that of the AD group(P<0.05).Conclusion:BDNF plays a very important role in protective brain neurons and maintenance their normal function,demonstrates the attractive prospect in the AD treatment.EA may obviously prevent the AD mouse to the learning and memory ability.Its possible mechanism promising:(1) Enhance the contents of BONF,which can promote the regeneration of brain neurons.(2) Enhance the contents of BDNF,TrkB in order to stimulate the CREB which is in karyon,thus enhances BDNF in the hippocampus from the secretion,protects brain neurons.
