| Objective:Based on the conclusion that the Qixian Tongluo Decoction is enable to restore the neurological function of the MCAO rats,this experiment aims to showcase the influence of the Qixian Tongluo Decoction on the MCAO rats’synaptic remodeling of the BDNF/TrkB/CREB signaling pathway and explore the rationaleMethods:Taking the MCAO rat model as the research object,the Chinese medicine compound Qixiantongluo Decoction and its blocker K252a were intervened based on the theory of Chinese medicine master Liu Zuyi’s "brain marrow Yangshengyinchang";Berdson score,BWT neural function score,immunohistochemistry,transmission electron microscopy,Western Blot and other modern biological methods,dynamic detection of rat nerve function after cerebral infarction,synaptic remodeling in the peripheral area of cerebral infarction,BDNF,TrkB,CREB,p-CREB protein expression level;study the ultrastructural basis of Qixian Tongluo Decoction in promoting neurological function recovery after cerebral infarction and the molecular mechanism of the prescription on synaptic remodelingResults:1.General conditions:Compared with those in the sham operation group,the rats in the Chinese-medicine and the citicoline groups both are improved significantly on their mental state,body reaction,fur colour and appetite 14 days later after the operation2.Behavioral score:(1)Berdson score:Since the surgery taken,the scores of the rats in the model group have gradually increased(P<0.01);the scores of the rats in the traditional Chinese medicine group have begun to decrease gradually 3 days later(P<0.05);the scores of the rats in the alkali group have gradually decreased 7 days later(P<0.01);(2)BWT score:Since the surgery taken,the scores of the rats in the traditional Chinese medicine group have gradually increased 3 days later(P<0.01);the scores of the rats in the Chinese medicine group have increased compared with the rats in the solvent group,the model group,the K252a+Chinese medicine group,and K252a group(P<0.05)3.Relevant indicators of synapse ultrastructure:(1)PSD thickness:The PSD thickness of the rats in the Chinese medicine group significantly increases compared with the rats in the sham-operation group,the model group,the K252a group,K252a+Chinese medicine group,and the solvent group(P<0.05)(2)Width of synaptic space:The width of synaptic space of the rats in the traditional Chinese medicine group significantly decreases compared with the model group(P<0.05)and the citicoline group(P<0.05);(3)Length of synaptic active area:The length of the synaptic active area of the rats in the Chinese medicine group significantly increases compared with other groups(P<0.05);the length of the synaptic active area of the rats in the citicoline group significantly increases compared with that of the K252a+Chinese medicine group(P<0.05)4.Immunohistochemistry:(1)PSD-95:The average optical density of PSD-95 of the rats in the traditional Chinese medicine group is higher than that of the other groups(P<0.05);the average optical density of PSD-95 of the rats in the citicoline group is higher than that of the model group,the K252a group,the K252a+Chinese medicine group,and the solvent group(P<0.05);(2)SYN:the average optical density value of SYN of the rats in the Chinese medicine group significantly increases compared with the other groups(P<0.05);the average optical density of SYN of the rats in the choline group is higher than that of the model group,K252a group,K252a+Chinese medicine group,and solvent group(P<0.05)5.Westernblot:(1)BDNF protein expression level:the BDNF protein expression level of the rats in the Chinese medicine group increases compared with that of the K252a group,K252a+Chinese medicine group,and citicoline group;(2)the p-CREB protein expression level of the rats in the Chinese medicine group increases compared with that of the K252a+Chinese medicine group,citicoline group,and solvent group(P<0.05)Conclusion:1.Qixian Tongluo Decoction promotes the recovery of neurological function in MCAO model rats,whose effect is better than citicoline;2.Qixian Tongluo Decoction promotes the recovery of neurological function by changing the synaptic ultrastructure and improving the synaptic remodeling of the rats;3.The BDNF/TrkB/CREB signaling pathway could be one of the main mechanisms which determines the promotion of Qixian Tongluo Decoction on the synaptic remodeling of neuronal cells. |
