Effects Of Cluster Needling Of Scalp Point Therapy Combined With Environmental Stimulation On Apoptosis And Expression Of Proteins Wnt1 And β-catenin In Neonatal Rats With Hypoxic-Ischemic Brain Damage

Objective:To explore the effects of cluster needling of scalp point therapy combined with environmental stimulation on neurological function,apoptosis in cerebral cortex and the expression of related proteins Wntl and β-catenin in neonatal rats with HIBD,expecting to use cluster needling of scalp point therapy combined with environmental stimulation to slow down cell apoptosis in clinical practice to provide a more valuable experimental basis for the treatment of HIBD.Methods:Ninety 7-day-old Wistar rats were randomly divided into sham operation group,model group,cluster needling of scalp point therapy group,environmental stimulation group and cluster needling of scalp point therapy combined with environmental stimulation group(combination group),and at 12h,24h and 7d were again divided into 3 time subgroups with 6 animals in each group.The HIBD modeling was carried out with reference to the modified Rice method.The rats in the sham operation and model groups were not given any treatment.The cluster needling of scalp point therapy group received cluster needling of scalp point therapy,the environmental stimulation group received environmental stimulation,and the cluster needling of scalp point therapy combined with environmental stimulation group received environmental stimulation at the same time during the long duration of cluster needling of scalp point therapy.At each corresponding time point,the modified Bederson score was used to evaluate the neurological function of the rats in each group.After the brain tissue was collected,TUNEL method was used to detect neuronal apoptosis in cortex,Western blot method was used to detect the expression of Wntl in brain tissue,and immunohistochemical method was used to detect the expression of β-catenin in brain tissue.Results:1.Bederson score:Compared with the sham operation group,the Bederson score of newborn rats in other groups at each time point showed extremely significant difference(P<0.01);Compared with model group,Bederson score of all treatment groups at each time point decreased significantly(P<0.01,P<0.05);Except for the 12h time point,at the 24h and 7d time points,the Bederson score of the combined group was significantly lower than that of the cluster needling of scalp point therapy group and the environmental stimulation group,and the difference was significant(P<0.05).2.Apoptosis of cerebral cortex:Except for the sham operation group,the positive cell apoptosis index at each time point in the remaining groups showed an upward trend;And compared with the sham operation group,the positive cell apoptosis index of the other groups at each time point was significantly different(P<0.01);Compared with the model group,the positive cell apoptosis index in each treatment group was significantly decreased at each time point,and the difference was significant(P<0.01,P<0.05);Compared with the cluster needling of scalp point therapy group and the environmental stimulation group,the positive cell apoptosis index of the combination group decreased significantly at each time point,and the difference was also significant(P<0.05).3.Expression of Wntl and β-catenin protein in brain tissue:Except for the sham operation group,the expression of Wntl and β-catenin protein in the remaining groups at each time point showed a general upward trend,which was consistent with the trend of apoptosis.And compared with the sham operation group,the other groups had extremely significant differences in Wntl and β-catenin protein expression at each time point(P<0.01);Compared with the model group,the protein expressions of Wntl and β-catenin in each treatment group increased significantly at each time point,and the difference was extremely significant(P<0.01);At each time point,the expression levels of Wntl and β-catenin proteins in the combination group were significantly higher than those in the cluster needling of scalp point therapy group and the environmental stimulation group,and the difference was also significant(P<0.01,P<0.05).Conclusion:1.Cluster needling of scalp point therapy,environmental stimulation and their combination can effectively promote the repair of damaged nerve function of HIBD newborn rats;can effectively improve neuronal apoptosis in the cerebral cortex of HIBD newborn rats;can increase the expression of Wntl and β-catenin proteins in the brain tissue of HIBD neonatal rats,and the curative effect of the combined group is better than the single cluster needling of scalp point therapy and environmental stimulation.2.Cluster needling of scalp point therapy combined with environmental stimulation may reduce apoptosis by increasing the expression of Wntl andβ-catenin proteins in brain tissue of HIBD newborn rats.
