| Objective:In this study,the villi tissue of patients with early embryo loss will be detected to describe the expression of Ng BR,VEGF,and P-Akt,and further analyze their possible roles in early pregnancy loss and explore their associations.It provides a theoretical basis for the formation of blood vessels in early pregnancy embryos.Methods:1.The subjects were from patients who underwent induced abortion in our hospital from July to December 2020.According to the reasons for the abortion,the patients were divided into a case group with early pregnancy loss and a control group with voluntary termination of pregnancy,with 25 cases in each group.2.The expression of Ng BR,VEGF and P-Akt protein in the villus tissue of the case group and the control group was detected by immunohistochemical method SP method;3.The expression levels of Ng BR,VEGF and P-Akt in the villus tissue of the case group and the control group were detected by real-time fluorescence quantitative PCR method;4.The measurement data were tested for normality and homogeneity of variance between groups.The mean and standard deviation were used for description,and the t test was used for comparison between groups.The exponentially distributed data were log-transformed before further analysis.Pearson correlation was used to test the correlation,and the correlation strength was described according to the magnitude of the correlation coefficient.In the above inspections,the inspection level α is set to 0.05.Results:1.Immunohistochemical SP test results1.1 Ng BR protein is mainly expressed in the cytoplasm and cell membrane of villus tissue,and the staining score of this protein in the case group was lower than that in the control group(2.06±0.67 VS 4.78±0.79,P<0.05);1.2 VEGF protein is mainly expressed in the cytoplasm and cell membrane of villi,and the staining score of this protein in the case group was lower than that in the control group(1.79±0.57 VS 4.64±0.82,P<0.05);1.3 P-Akt protein was mainly expressed in the cytoplasm of villus tissue,and the staining score of this protein in the case group was lower than that in the control group(1.68±0.59 VS 4.64±0.96,P<0.05).2.RT-PCR test results and correlation2.1 Compared with the control group,the relative expression of Ng BR m RNA in the villus tissue of the case group was lower(1.48±1.35 in the case group and 2.75±1.78 in the control group),and the difference was statistically significant(P<0.05);2.2 Compared with the control group,the relative expression of VEGFm RNA in the villi tissue of the case group was lower(1.67±1.35 in the case group and 2.59±1.97 in the control group),and the difference was statistically significant(P<0.05);2.3 Compared with the control group,the relative expression of P-Akt m RNA in the villus tissue of the case group was lower(1.56±1.18 in the case group and 2.85±1.09 in the control group),and the difference was statistically significant(P<0.05);2.4 Pearson correlation analysis was used to test the correlation of m RNA expression in patients,and it was found that the expressions of the above three were positively correlated(r=0.782,0.753,0.649,P<0.05).2.4 Pearson correlation analysis was used to test the correlation of m RNA expression in patients,and it was found that the expressions of the above three were positively correlated(r=0.782,0.753,0.649,P<0.05).Conclusion:1.There were expressions of Ng BR,VEGF and P-Akt proteins in the villus tissue of both the case group and the control group;2.The protein expression and m RNA expression of Ng BR,VEGF and P-Akt m RNA in the villus tissue of the case group were lower than those of the control group;3.The expression of Ng BR,VEGF and P-Akt m RNA in the villi tissue of the case group was positively correlated,suggesting that the low expression of the three is involved in the pathogenesis of early pregnancy loss. |
