| Objective:The expression of FOXO1 and VCAM-1 in villus tissue of normal early pregnancy and early pregnancy loss patients was detected,and the correlation between Fox O1 and VCAM-1 in early pregnancy loss was discussed.Methods:1.during August 1 2020-January 30 2021,we were collected the villus tissues of 50 patients who voluntarily underwent induced abortion in the gynecological outpatient department in the Second Hospital of Shanxi Medical University.One group was the case group,which included 25 patients with early pregnancy loss and the other group was a control group,which included 25 patients with normal early pregnancies.2.The expression of FOXO1 and VCAM-1 in villus tissue of early pregnancy loss group and normal early pregnancy group was detected by immunohistochemical method.The expression of FOXO1 and VCAM-1 mRNA in the villi were detected by qPCR.3.Statistical method SPSS26.0 software was used to conduct statistical analysis.The homogeneity of variance and normality of all the data obtained from the experiment were tested first.If the data did not conform to the normal distribution,it was necessary to convert the data logarithmically before analysis.For case group and control group,independent sample t test was used for comparison of the same index between the two groups and Pearson analysis was used for the correlation between various indicators.P<0.05 was considered statistically significant.Results:1.There was no statistical significance in the comparison of the general information of the early pregnancy loss group and the normal early pregnancy group,including age,gestational age,gestational order,birth order,BMI and blood pressure(P > 0.05).2.Immunohistochemical results FOXO1 and VCAM-1 scores(2.67±1.39 and 2.73±1.12,respectively)in the group with early pregnancy loss were lower than those in the normal group(4.37±1.13 and 4.73 ± 1.23,respectively).There were statistically significant differences in all indicators between the two groups(P < 0.05).3.TheqPCR results The relative expression levels of FOXO1 and VCAM-1 in villus tissues of the early pregnancy loss group(0.77±1.46,0.43±0.17,respectively)were lower than those of the normal early pregnancy group(1.61 ± 0.38,1.23 ± 0.36,respectively).The independent sample t test for each index of the early pregnancy loss group and the normal early pregnancy group showed P < 0.05,suggesting that the expression differences of FOXO1 and VCAM-1 between the early pregnancy loss group and the normal early pregnancy group were statistically significant.Results Pearson correlation analysis showed that FOXO1 was positively correlated with VCAM-1,and the difference was statistically significant(P < 0.05).Conclusion:1.FOXO1 and VCAM-1 were expressed in villi in early pregnancy.2.The expression of FOXO1 and VCAM-1 in villus tissue of the early pregnancy loss group was lower than that of the normal early pregnancy group.3.FOXO1 and VCAM-1 were correlated in the villous tissue of the early pregnancy loss group,suggesting that FOXO1 and VCAM-1 may be involved in the occurrence and development of early pregnancy loss.However,the molecular mechanism is still unclear,providing a new idea for the clinical research on the pathogenesis of early pregnancy loss. |
