Research purposes:The theory of poison and blood stasis interlocking is similar to the mechanism of vascular restenosis caused by inflammatory reaction after percutaneous coronary intervention(PCI).Necrosis factor-related receptor factor 2/nuclear transcription factor-κ B/myeloid differentiation factor 88(TRAF2/NF-κ B/My D88)signaling pathway to observe the effect of Jiedu Huoxue Formula on systemic and local inflammatory responses in SD rats after balloon injury To explore the mechanism of Jiedu Huoxue Formula in intervening inflammatory response,inhibiting the proliferation and migration of vascular smooth muscle cells,and preventing vascular restenosis,so as to provide a scientific basis for the clinical treatment of vascular restenosis after PCI by Jiedu Huoxue Formula.Research method:1.Modeling: In this experiment,8-week-old male Sprague Dwaley(SD)rats were selected to establish a thoracic aortic intimal hyperplasia model.2.Grouping and administration: 72 SD rats were divided into 6 groups according to the random number method,namely the sham operation group(group S,distilled water),the modeling group(group M,distilled water),and the aspirin group(group P,equivalent).In aspirin enteric-coated tablets 15mg/Kg/d),Jiedu Huoxue Formula low-dose group(JL group,equivalent to crude drug concentration 2.13g/Kg/d),Jiedu Huoxue Formula medium-dose group(JM group,equivalent to crude drug concentration4.26g/Kg/d),Jiedu Huoxue Formula high-dose group(JH group,equivalent to crude drug concentration 8.53g/Kg/d).There were 12 rats in each group,and the rats were weighed every 14 days.The dosage regimen was adjusted according to the change of body weight.The total number of administration days was 28 days.The animals were sacrificed in two batches 14 days and 28 days after modeling.3.Detection methods and indicators: 14 and 28 days after operation,the rats were sacrificed in two batches,the abdominal aorta of each group was collected,and the levels of serum IL-1 β and TNF-α were detected by Elisa method;The protein expression levels of IL-1β、TNF-α、Cyclin D1、 Cyclin E、TRAF2、My D88 and NF-κB p65 were detected by Western blot.Research result:1.Comparison of serum inflammatory factors: 14 days after operation,compared with group S,the levels of IL-1 β and TNF-α in group M increased(P<0.05 or P<0.01);compared with group M,JL,JM and The content of IL-1βin the JH group was decreased(P<0.05 or P<0.01),and the content of TNF-α in the P and JH groups was decreased(P<0.05 or P<0.01).Compared with the P group,there was no significant difference in the levels of IL-1βand TNF-α in the JL,JM and JH groups(P>0.05).On the 28 th day after operation,compared with group S,the levels of IL-1βand TNF-α in group M were significantly increased(P<0.05 or P<0.01);The content of TNF-αwas significantly decreased(P<0.05 or P<0.01),and the content of IL-1β in the JL and JM groups was significantly decreased(P<0.01);compared with the P group,the content of IL-1β in the JH group was significantly decreased(P<0.05).2.Comparison of vascular inflammatory proteins: 14 days after operation,compared with group S,the levels of IL-1βand TNF-α in group M were increased(P<0.01);The levels of IL-1βand TNF-α were significantly decreased(P<0.05 or P<0.01);compared with the P group,there was no significant difference in the TNF-αlevel in the JH group(P>0.05).On the 28 th day after operation,compared with group S,the levels of IL-1βand TNF-α in group M were increased(P<0.01);compared with group M,the levels of IL-1βand TNF-α in groups P,JM and JH were significantly increased.Compared with the P group,there was no significant difference in the levels of IL-1βand TNF-α in the JH group(P>0.05).3.Comparison of cyclin: 14 days after operation,compared with group S,the levels of Cyclin D1 and Cyclin E in group M were significantly increased(P<0.01);compared with group M,the levels of Cyclin D1 and Cyclin E in groups P,JM and JH were significantly increased.Compared with the P group,there was no significant difference in the levels of Cyclin D1 and Cyclin E between the JM and JH groups(P>0.05).On the28 th day after operation,compared with group S,the levels of Cyclin D1 and Cyclin E in group M were significantly increased(P<0.01);compared with group M,the levels of Cyclin D1 and Cyclin E in groups P,JL,JM and JH were significantly decreased(P<0.05 or P<0.01);compared with the P group,there was no significant difference in the levels of Cyclin D1 and Cyclin E between the JM and JH groups(P>0.05).4.Comparison of vascular signaling pathway proteins: 14 days after surgery,compared with group S,the levels of TRAF2、My D88 and NF-κB p65 in group M were significantly increased(P<0.01);The levels of TRAF2、My D88 and NF-κB p65 in the group were significantly decreased(P<0.05 or P<0.01);compared with the P group,there was no significant difference in the levels of TRAF2、My D88 and NF-κB p65 in the JH group(P>0.05).28 days after operation,compared with group S,the levels of TRAF2、My D88 and NF-κB p65 in group M were significantly increased(P<0.01);compared with group M,the levels of TRAF2、My D88 and NF-κB p65 in group P and JH Compared with the P group,there was no significant difference in the levels of TRAF2、My D88 and NF-κB p65 in the JH group(P>0.05).Analysis conclusion:1.The thoracic aortic intimal hyperplasia model established by balloon injury of rat thoracic aorta can provide an experimental basis for future research on vascular restenosis.2.Jiedu Huoxue Formula can effectively reduce the content of IL-1β and TNF-α in serum and protein expression in vascular tissue of SD rats after balloon injury,and reduce the inflammatory response.3.Jiedu Huoxue Formula can reduce the protein expression levels of Cyclin D1 and Cyclin E in the vascular tissue of SD rats after balloon injury by regulating cell cycle protein,and achieve the effect of inhibiting the proliferation of vascular smooth muscle cells.4.Jiedu Huoxue Formula can antagonize the inflammatory response by interfering in the activation and conduction of the TRAF2/NF-κ B/My D88 signaling pathway,thereby inhibiting the proliferation,migration and intimal hyperplasia of vascular smooth muscle cells in SD rats after balloon injury,and ultimately preventing Vascular restenosis.
