ObjectiveAcute coronary syndrome(ACS),which is caused by rupture of a coronary arterial atherosclerotic plaque,is a serious threat to human life and health.Atherosclerosis is considered as a "chronic inflammatory response".With the progress of the study,we found that angiogenesis is often accompanied with the inflammatory response within the plaque,the number of neovascularization is closely related to the stability plaque.Therefore,how to inhibit the angiogenesis within the plaque is a big question to prevent and treat the atherosclerosis.VEGF as a specific vascular growth factor,has become the main target of treating atherosclerosis.Wenyang-Huoxue-Jiedu-Formula(WHJF),which is an empirical formula used to treating coronary heart disease(CHD)for many years,showed favorable efficacy.To study the mechanism of WHJF treating the thin-cap fibro atheroma plaques(TCFA),a series of experiments and clinical trials was designed in this paper.MethodsThe ApoE-/-rats were used to made the AS model by feeding with high fat diet for 13 weeks.Then,they were randomly divided into four groups:the control group,low-dose WHJF group,middle-dose WHJF group,high-dose WHJF group,which were respectively treated with normal saline,WHJF(14.08g · kg-1,28.16g · kg-1,56.32g · kg-1)a day for another 13 weeks.The expressions of VEGF,VEGFR1 and VEGFR2 in the aorta of rats were detected by western blot respectively.A total of 40 patients with non-ST-segment elevation acute coronary syndrome were randomly divided into two groups:routine treatment group and traditional Chinese medicine treatment group(20 cases).The course of treatment was 2 weeks.The patients’ TCM syndrome score,TCM symptoms and platelet agglutination test(PAgT)were observed to evaluate the clinical efficacy of WHJF.Results:There was no significant difference in the expression of VEGF,VEGFR1 and VEGFR2 in the aorta of the AS model mice between the control group and the WHJF groups(P>0.05).Compared with the conventional treatment group,the traditional Chinese medicine treatment group can not only improve the assessment of TCM syndrome effectively,but also decreased platelet aggregation rate.ConclusionWHJF has no effect on the expression of VEGF,VEGFR1 and VEGFR2 in VEGF/VEGFRs signaling pathway of AS model mice.However,combined with the results of previous in vitro experiments,we deduce that WHJF may adjust pERK1/2 signal molecules to achieve the inhibition of angiogenesis.In addition,WHJF can not only improve the symptoms of patients,but also play the role of anti-platelet aggregation. |
