Objective: In this study,differences in GAS5 expression levels in peripheral blood of patients in the stent restenosis group(ISR)and the non-restenosis group(N-ISR)after Percutaneous coronary intervention(PCI)were analyzed to study the correlation between GAS5 and in-stent restenosis.GAS5 was used as a New biological markers and therapeutic targets for in-stent restenosis provide new research directions.Methods:Among the patients with previous stent placement admitted to the Department of Cardiology,Affiliated Hospital of Jining Medical University from April 2019 to December 2020,the patients were divided into the restenosis group and the non-restenosis group according to the presence of stent restenosis after angiography review.Twenty-six patients in the stent restenosis group were selected continuously(ISR,n=26).Twenty-six patients without restenosis group were randomly selected(N-ISR,N =26).First,peripheral blood of selected patients was collected to extract monocytes from the peripheral blood,and then total RNA from the above-mentioned cells was extracted.Real-time fluorescence quantitative polymerase chain reaction was used to detect the expression level of GAS5 in the two groups of patients.Finally,whether the difference was statistically significant was evaluated.Univariate and multivariate logistic regression analysis was used to study the independent risk factors of ISR patients.Finally,the area under the ROC curve was used to evaluate the potential of GAS5 as a molecular marker of ISR.Results: 1.The expression of GAS5 in the ISR group was lower than that in the N-ISR group(1.090.45vs1.690.60),and the difference was statistically significant(P < 0.05).2.Multivariate logistic regression analysis was conducted on statistically significant smoking,GAS5 and diabetes in the ISR group and the N-ISR group,and the results showed that GAS5,smoking and diabetes were independent risk factors for ISR.3.ROC curve analysis is adopted to calculate the Areaunderthecurve(AUC)to evaluate whether the relative expression of GAS5 has predictive value for ISR.The results showed that the AUC of GAS5 was0.804,(95%CI 0.693,0.915).When the Youden index was 0.423,the sensitivity and specificity of GAS5 in the diagnosis of ISR were 80.8%and 61.5%.Conclusion:1.Gas5 with low expression level is an independent risk factor for ISR patients,which may be a new biomarker for predicting ISR.2.The decreased expression level of GAS5 in ISR patients may promote the proliferation and migration of smooth muscle cells through a series of mechanisms,which may be one of the occurrence mechanisms of ISR.
