| This subject adopts single factor investigation,orthogonal design and response surface methodology were used to optimize the extraction process of Pfaffia glomerata and Pfaffia glomerata compound(the compound is an independent prescription,including 9 g Pfaffia glomerata,9 g Lycium barbarum L.,9 g Mulberry,5 g flowers of the Dolichos lablab L.and 5 gflowers of the Citrus aurantium L.).The anti-fatigue and regulatory immunity of Pfaffia glomerata and its compound were investigated.On this basis,Pfaffia glomerata and its compound preparations were developed to expand the application range of Pfaffia glomerata.The specific research methods and results are as follows:1.Research on the extraction technology of Pfaffia glomerataTaking the extraction amount of oleanolic acid,ecdysterone and extract as indexes,the extraction process of Pfaffia glomerata was optimized by single factor and orthogonal design:80%ethanol 1:10(g·m L-1)was refluxed for 1 h at 100°C and extracted twice.Among them,the extraction amount of oleanolic acid was 22.06μg·g-1,the extraction amount of ecdysterone was 5.18 mg·g-1,and the extraction rate of extract was 54.49%.2.Research on the extraction technology of Pfaffia glomerata compoundTaking the extraction amount and extract amount of total polysaccharides,total saponins,ecdysone,rutin and resveratrol as indexes,the extraction process of Pfaffia glomerata compound was optimized by single factor,hierarchical analysis and response surface design method:water 1:12(g·m L-1)decoction,extraction 140 min,extraction twice;the extraction rate of extract was 51.22%,the extraction amount of total polysaccharides was 170.577 mg·g-1,the extraction amount of total saponins was52.091 mg·g-1,the extraction amount of ecdysone was 1.236 mg·g-1,the extraction amount of rutin was 1.262 mg·g-1,and the extraction amount of resveratrol was0.0769 mg·g-1.3.Research on anti-fatigue and regulating immunity of Pfaffia glomerata and its compoundIn this paper,the anti-fatigue effect of Pfaffia glomerata and its compound was investigated by swimming time,liver/muscle glycogen content,serum LA and BNN content in mice.The effect of Pfaffia glomerata and its compound on regulating immunity was investigated by organ/body weight ratio,Ig G,Ig M,Ig A immunoglobulin content,Con A-induced lymphocyte transformation rate,NK cell activity;DTH value,serum HC50content and hemolytic plaque number in immunocompromised mice.Research on the anti-fatigue and immunoregulatory effects of Pfaffia glomerata:Compared with the blank group,the high-dose Pfaffia glomerata group significantly prolonged the swimming time of mice under load,and significantly increased the glycogen content of liver and muscle(P<0.05).Pfaffia glomerata reduced BUN and serum LA activity in mice(P>0.05).The results showed that Pfaffia glomerata had anti-fatigue effect.The immunocompromised model of mice was used to investigate the effect of promoting immunity.Compared with the model group,Pfaffia glomerata and the positive control group increased the contents of Ig G,Ig M and Ig A immunoglobulins in mice(P>0.05).Pfaffia glomerata high dose group and positive control group significantly increased Con A-induced lymphocyte transformation rate and NK cell activity in mice(P<0.05).Normal mice were used to verify the immunoregulatory effect.Compared with the blank group,the DTH and HC50of mice in the high-dose group and the positive control group were significantly increased.(P<0.05).Pfaffia glomerata increased the number of hemolytic plaque in mice(P>0.05).The results showed that Pfaffia glomerata had immunoregulatory effects on cellular immunity and nonspecific immunity in mice.Research on the anti-fatigue and regulating immunity of Pfaffia glomerata compound:Compared with the blank group,Pfaffia glomerata compound middle and high dose group significantly prolonged the load swimming time of and HC50of mice in the medium-and high-dose groups of Pfaffia glomerata compound and the positive control group were significantly increased(P<0.05)Pfaffia glomerata compound increased the number of hemolytic plaque in mice(P>0.05).The results showed that Pfaffia glomerata compound had immunoregulatory effects on humoral immunity,cellular immunity and nonspecific immunity in mice.4.Prescription optimization and process research of Pfaffia glomerata chewing gumWith hardness,elasticity,viscosity,chewiness and sensory evaluation as indexes,single factor and orthogonal design were used to investigate the preparation formula of Pfaffia glomerata chewing gum:35%gum base,40%xylitol,7%freeze-dried powder of Pfaffia glomerata extract,1%green tea powder,0.5%matcha powder,25%glycerol,0.4%citric acid,0.8%Peppermin oil and 0.5%Achyrocline satureioides.5.Preparation technology and quality control of Pfaffia glomerata compound granulesWith molding rate,particle size,water solubility,dissolution rate,angle of repose,moisture absorption rate and sensory evaluation as indexes,the preparation prescription of Pfaffia glomerata compound granules was investigated by single factor and response surface methodology:50%freeze-dried powder of Pfaffia glomerata compound extract,12.5%soluble starch,37.5%cyclodextrin,and 89%ethanol 16%,mixed well,made soft materials,granulated,dried at 60°C for 1 h,and graded whole granules.According to the requirements of 2020’Chinese Pharmacopoeia’granules,the inspection of Pfaffia glomerata compound granules meets the requirements of 2020’Chinese Pharmacopoeia’granules. |
