| Objective: To observe the changes of IL-17 A and autophagy of bronchial fibroblasts in lung tissues of COPD patients,and to speculate the relationship between the expression of IL-17 A and autophagy of bronchial fibroblasts with airway remodeling.Methods:1.Experimental grouping and sampling: lung tissue samples of patients undergoing pulmonary lobectomy in thoracic surgery in our hospital were selected,including 10 cases without COPD(control group)and 10 cases in COPD group(6cases met the clinical diagnosis of COPD and 4 cases met the pathological changes of COPD).All specimens were taken from 5cm far from the lesion edge.2.HE staining observed the pathological pathological changes of lung tissue.3.Masson staining observed collagen deposition and airway remodeling of lung tissues.4.IL-17 A was measured by immunohistochemistry to observe whether there was any difference in IL-17 A expression between the two groups.5.The expression of LC3Ⅱ/Ⅰ protein in lung tissue was measured by Western blot(WB).6.Immunofluorescence triple staining was used to observe autophagy of bronchial fibroblasts: fibroblast specific marker protein(FSP1 and vimentin),autophagy specific marker protein(LC3B).Results:1.HE staining results: In COPD group,inflammatory cells(such as neutrophils,lymphocytes,etc.)were seen in the small airway infiltration,some epithelial cell degeneration and necrosis,some connective tissue and smooth muscle tissue hyperplasia,some alveolar septum discontinuous and alveolar cavity enlargement.2.Masson staining results: Compared with the control group,collagen deposition was obvious in the COPD group(P< 0.05).3.Immunohistochemical results: Compared with the control group,IL-17 A expression was increased in COPD patients,and the difference was statistically significant(P< 0.05).4.Immunofluorescence results: Bronchial fibroblasts were successfully located,and LC3 B fluorescence expression was enhanced in the COPD group compared with the control group,with statistically significant difference(P< 0.05)5.WB results: Compared with the control group,the increased expression of LC3 Ⅱ/ Ⅰprotein in lung tissues of COPD group was statistically significant(P< 0.05).Conclusions: 1.Airway inflammation and remodeling were found in lung tissues of COPD patients again。2.IL-17 A expression was increased in COPD group.3.Autophagy is enhanced in lung tissue and bronchial fibroblast in COPD patients.It can be inferred that the enhancement of bronchial fibroblast autophagy may be involved in airway remodeling in COPD. |
