Objective By detecting the differences in expression level of autophagy relevant genes and protein LC3 and p62,as well as the quantity and apoptosis of the CD8+T lymphocyte in peripheral blood of Chronic Obstructive Pulmonary Disease(COPD)patients in catabasis,so as to explore the correlation research between the autophagy level and the quantity of CD8+T lymphocyte.To explore the causes of the increase of CD8+T cells in patients with COPD in catabasis,which it is helpful to clarify the pathogenesis of COPD.Methods There are three groups,which contain COPD group,smoking with normal lung function group(SN group),and non-smoking with normal lung function group(NSN group),each group contains 30 cases.Using FCW to detect the ratio of CD8+T cells,CD8+effector T cells and CD8+memory T cells as well as the apoptosis rate of them.Sorting out CD8+T cells in Trizol by flow cytometry(FCW),using real-time quantitative PCR(qRT-PCR)and Western Blot to detect the expression level of CD8+T lymphocytes autophagy relevant genes LC3,p62 in peripheral blood.Results 1.Flow cytometry detection shows:(1)The quantity of CD8+T cells in COPD group(32.33±4.23%)was higher than the other two groups,and the results were statistically significant(P<0.05).The quantiiest of both CD8+effector T cells(25.63±8.57%)and CD8+memory T cells(11.94±5.77%)were higher in COPD group than the other two groups(P<0.05).The number of CD8+T cells,CD8+effusive T cells and memory T cells in the SN group were higher than those in NSN group(p<0.05).(2)The apoptosis rate(23.51±6.11%)of CD8+T lymphocytes in COPD group was lower than those in the other two groups(P<0.05),while the rate in SN group was lower than that in NSN group(p<0.05).2.there was no significant difference among the groups in expression level of gene LC3(P>0.05).However,the expression levels of gene P62 was higher in COPD group than the SN normal group(p<0.05),while the level in SN group was higer than the NSN group(p<0.05).3.The expression level of LC3Ⅱ/Ⅰin COPD group was higher than those in SN group(p<0.05),while the level in SN group was higher than that in NSN group.As to P62,the expression level in COPD group was higher than that in SN group(p<0.05),while the SN group was higher than the NSN group(p<0.05).4.Pearson correlation analysis showed that within the COPD group,the expressing level of LC3Ⅱ/Ⅰhas a positive correlation with the quantity of CD8+T cells(r=0.572,p<0.05),and CD8+effector T cells(r=0.672,p<0.05),and CD8+memory T cells(r=0.739,p<0.05).There was no correlation between the expression leve of LC3Ⅱ/Ⅰand the apoptosis rate of CD8+T cells.(2).The expression level of P62 had a positive correlation with the quantity of CD8+T cells(r=0.613,p<0.05),and CD8+effector T cells(r=0.681,p<0.05),and CD8+memory T cells(r=0.745,p<0.05).The expression level of P62 was not significantly correlated with the apoptosis rate of CD8+T cells.5.regression analysis showed that within the COPD group,the linear connection between LC3Ⅱ/Ⅰ/GAPDH(‰)and the quantity of CD8+T cells,and the quantity of CD8+effective T cells,and the quantity of CD8+memory T cells were all statistically significant(P<0.001).(2)the linear connection between P62/GAPDH(‰)and the quantity of CD8+T cells,and the quantity of CD8+effective T cells,and the quantity of CD8+memory T cells were all statistically significant(P<0.001).Conclusions 1.The quantity of CD8+T cells,CD8+effector T cells and CD8+memory T cells in peripheral blood of COPD patients in stable phase all increased significantly,while the apoptosis rate of them were lower.2.The expressing level of autophagy correlation protein LC3Ⅱ/Ⅰand P62 were significantly increased in the stable phase of COPD patients.3.The increased autophagy level and the increased quantity of CD8+T cells had a positive correlation and linear connection,which suggest that the cell autophagy join in the pathogenesis of COPD.
