Objective:To study the effect of IFNL3/4 gene polymorphism on the therapeutic response of HARRT in AIDS patients,The genotypes of IFNL3/4 gene loci rs28416813,rs12971396 and rs4803221 were detected,and genotype frequencies and allele frequencies were compared between the two groups with different response to HARRT.After that,the correlation between SNPs of IFNL3/4 gene locus and the curative effect of HARRT in AIDS patients were comprehensively analyzed.Methods:1.All the subjects in this study are from HIV-infected/AIDS patients who visited Ruikang Hospital affiliated to Guangxi University of Chinese Medicine from January 2011 to January 2020.According to the modern medical diagnostic criteria and the inclusion criteria and exclusion criteria of this study,137 HIV infected/AIDS patients were selected by random sampling method.And their clinical data and whole blood were collected.Genomic DNA was extracted from anticoagulation peripheral blood of each subjects.Oligo 7 Primer software was used to design specific external primers for each site,and two pairs of primers-polymerase chain reaction(PCR-CTPP)were used to amplify the target genes of IFNL3/4rs28416813,rs12971396 and rs4803221,and 5%of the positive amplification products were purified and sent for sequencing,and the sequencing results were analyzed by Bioedit for genotyping.2.According to the evaluation standard of efficacy of immunological treatment in Chinese AIDS Diagnosis and Treatment Guide(2021 edition),the CD4~+T cell counts increased by 100/μL compared with the baseline after one year of ART treatment was classified as the valid group,otherwise it was classified as invalid.The genotype and allele distribution of IFNL3/4 locus among different groups and their correlation with HAART treatment response were analyzed.3.Bivariate Logistic regression analysis was used to compare the influence of various factors on the treatment results,and odds ratio(OR)and 95%confidence intervals(95%CI)were calculated.The haplotype,allele frequency and linkage disequilibrium of genotyping were analyzed by SHEsis software.All the tests were bilateral,and the difference was statistically significant with P<0.05.Results:1.Gender,age,baseline CD4~+T cell counts,baseline CD8~+T cell counts and baseline CD4/CD8 ratio were not statistically significant between the valid group and inactive groups(man 74.40%vs74.50%,P=0.991;(38.31?14.71)years old vs(40.75?15.48)years old,P=0.361;(268.06?113.13)/μL vs(294.43?129.90)/μL,P=0.214;1001.41?454.31)/μL vs(1069.49?611.26)/μL,P=0.492;0.30?0.15 vs 0.37?0.39,P=0.211).2.SHEsis analysis showed that there was a high linkage imbalance between rs28416813 and rs12971396 loci of IFNL3/4.The linkage disequilibrium of rs4803221,rs28416813 and rs12971396 was low.3.Only GC genotype and CC genotype were detected in three loci of rs28416813,rs12971396 and rs4803221,no GG genotype was found.In univariate analysis,there was no correlation between the genotypes of rs28416813,rs12971396 and rs4803221 and the response to one year of TDF+3TC+EFV treatment(P=0.628、1.000、0.534,respectively).After adjusting for gender,age,baseline CD8 and baseline CD4/CD8,rs28416813-GC,rs12971396-GC and rs4803221-CC showed no correlation with HAART treatment response(P=0.824、0.732、0.591,respectively).There was no significant difference in allele frequencies of rs28416813,rs12971396 and rs4803221 between the valid group and inactive group(P=0.898、0.977、0.773,respectively).4.Sex stratified analysis showed that there was no significant difference in genotype frequency and allele frequency among the three loci(rs28416813,rs12971396 and rs4803221)between male and female of valid group and inactive groups(P>0.05).Conclusion:The gene polymorphisms of the three sites of rs28416813,rs12971396 and rs4803221 in IFNL3/4 gene region have no obvious correlation with the response of AIDS patients to TDF+3TC+EFV treatment.
