Efficacy And Mechanism Of Jiawei Erxian Decoction On Menopause-like Syndrome During Endocrine Therapy After Breast Cancer Surgery

Objective: In this study,the randomized controlled experiment method was used,combined with the emerging network pharmacology in recent years,to study the clinical efficacy of Jiawei Erxian Decoction in the treatment of climacteric syndrome of Chong-Ren disorder type during endocrine therapy of breast cancer,and to explore the efficacy of Jiawei Erxian Decoction in the treatment of climacteric syndrome.(Perimenopausal Syndrome,PMS)on the target pathway mechanism,and then provide theoretical support for further clinical research.Methods:Relevant case data were collected,and 60 patients who met the inclusion criteria were studied and randomly divided into control group and treatment group.,2consecutive courses of treatment.After the course of treatment,the Kupperman symptom score,TCM syndrome score,Karnofsky score changes and sex hormone levels were used to judge and compare the therapeutic effects before and after treatment.At the same time,combined with the related research methods of network pharmacology,the compounds and their targets with high activity in various traditional Chinese medicines of Jiawei Erxian Decoction were collected,and mapped with PMS disease-related targets,and their interaction networks were established and gene enrichment and analysis were carried out.Pathway prediction to explore the mechanism of action of Jiawei Erxian Decoction in the treatment of PMS.Results: After 4 weeks of treatment,the Kupperman symptom score in the treatment group was statistically significant(P<0.05),while the Kupperman score in the control group had no significant change(P>0.05).After 8 weeks of treatment,there were significant differences in Kupperman symptom score,TCM syndrome score and Karnofsky score in the treatment group(P < 0.001),while there was no significant change in the score in the control group(P > 0.05).There was no significant change in estradiol(E2)between the two groups before and after treatment(P>0.05).Through network pharmacology analysis,it was found that Jiawei Erxian Decoction mainly through IL-6,TNF,VEGFA,TP53,ESR1,CAT,IL-1β and other targets and specific biological pathways such as PI3K-Akt signaling pathway and HIF-1 signaling pathway It has many functions such as regulating estrogen in the body,maintaining the balance of osteoblasts and osteoclasts,improving breast tissue,and relieving the clinical symptoms of PMS patients.Conclusion: Modified Erxian Decoction has a definite curative effect on the treatment of Chong-Ren disorder breast cancer patients with climacteric syndrome during endocrine therapy,can reduce adverse drug reactions,and has obvious advantages in improving clinical symptoms and improving the quality of life of patients.Erxian Decoction can activate a large number of target genes,interfere with a variety of cellular biological processes and signaling pathways in the human body,and through partial and overall regulation,it has the characteristics of multi-level,multi-system and overall intervention in improving PMS.
