Objective:To evaluate the effects of acupuncture at Tianshu and Zusanli points on single and bilateral points on colon function and autonomic nervous balance in IBS-D rats by observing changes in colonic sensitivity, colonic slow wave discharge and heart rate frequency domain. To provide reference for optimizing and standardizing acupuncture and moxibustion for the treatment of IBS.Materials and Methods: Materials and methods: sixty male clean-grade Wistar rats weighing 200-250 g were randomly divided into four groups: a normal control group,a model group,an electroacupuncture unilateral Zusanli +Tianshu group, and an electroacupuncture bilateral Zusanli + Tianshu group.Water avoidance emergency method was used for modeling. After the end of modeling, the abdominal wall withdrawal reflex(AWR)semiquantitative scoring method was used to evaluate the model.Milliacupuncture at Zusanli acupoint about 0.5cm and Tianshu acupoint about 0.3cm was received from electroacupuncture diagnosis and treatment instruments with the following parameters: continuous wave,2Hz,1m A for 20 min once daily for 14 d.Grasping and immobilization were performed daily for the model and control groups by the same method, but electroacupuncture intervention was not given.Under anesthesia, after recording the ECG,the powerlab data acquisition and analysis system was used to record the discharge activity of the corresponding rectus abdominis muscle in rats under different distension pressures.A pair of electrodes was buried in the colon approximately 2 cm from the cecal end, with the electrodes approximately 3 mm apart and the line perpendicular to the long axis of the hollow viscera. Before colon sensitivity assay, standard II lead electrocardiograms were recorded using a powerlab data acquisition and analysis system.Results:(1)colonic pain sensitivity was significantly improved in IBS rats after treatment with both EA groups(P < 0.01);(2)After treatment in both EA groups, the colonic slow wave rhythm was well restored in IBS model rats, and the colonic slow wave frequency was significantly reduced(P < 0.001);(3)The LF,LF / HF of IBS rats were significantly decreased after treatment in both EA groups(P < 0.01);(4)There were no significant differences in colonic sensitivity, colonic slow waves,or frequency domain improvement between unilateral EA and bilateral EA.Conclusion: Both the unilateral EA group with Zusanli and Tianshu, and the bilateral EA group with Zusanli and Tianshu, can significantly improve the colon function and the balance of autonomic nerves in IBS model rats, but unilateral EA Zusanli and Tianshu can also be obtained by electroacupuncture on both sides of Zusanli and Tianshu.Preliminary tips: Electroacupuncture can use unilateral acupoints in the treatment of IBS,but clinical trials are still needed to verify whether it is consistent with the results of animal experiments.
