| Objective:In this study,heart rate variability,colorectal dilatation pain threshold,colonic electrogram and ELISA test results were used as objective indicators to observe the effects of unilateral and bilateral electro-acupuncture on the regulation of colonic function and autonomic homeostasis in rats with diarrhea-type irritable bowel syndrome,respectively,and to provide reference for clinical point selection.Methods:After the preparation of IBS-D model,the blank group was not given any intervention;after the model group was established,no intervention was given;the rats in the unilateral group were given electroacupuncture at Shangjuxu point,left and right alternately;the rats in the bilateral group were given electroacupuncture at bilateral Shangjuxu points,1 day/time,20 minutes each time,and the three groups were treated for 14 days.Normal feed and water were given during the period.The general condition,the number of loose stools and the total number of feces of each group of rats were observed and recorded every day,and the loose stool rate was calculated.Four groups of rats were given colorectal distension stimulation before and after treatment,and abdominal withdrawl reflex(AWR)score was used to record the score results.The electrocardiogram of rats was recorded by PowerLab experimental data acquisition and analysis system.The changes of low frequency(LF)signal,high frequency(HF)signal and LF/HF,colorectal stimulation-rectus abdominis reflex and colonic discharge activity were observed.Results:1.General situation of ratsThe rats in the blank group had normal eating and water intake,white hair,agile activity and few fighting phenomena.The rats in each model group had more defecation times,thin stool,fighting and biting,dry hair and light weight.After treatment,the diet and defecation of rats in the unilateral group and the bilateral group were improved.2.Fecal grading score and AWR score of ratsAt the end of modeling,the changes in fecal grading scores and their dilution rates in the IBS-D model rats were consistent with the characteristics of IBS-D.Before treatment,AWR was significantly higher in the model group,unilateral group and bilateral group compared with the blank group(P<0.05);after treatment,AWR score was significantly higher in the model group compared with the blank group(P<0.05),and significantly lower in the unilateral and bilateral groups compared with the model group(P<0.05);No significant difference between the unilateral and bilateral groups(P>0.05).3.Heart rate variability in ratsCompared with the null group,the model group showed an increase in low frequency(LF)signal,a decrease in high frequency(HF)signal and an increase in LF/HF(P<0.05);compared with the model group,the unilateral group showed a decrease in LF,an increase in HF and a decrease in LF/HF(P<0.05);the bilateral group showed a decrease in LF,an increase in HF,and a decrease in LF/HF(P<0.05).No significant difference between the unilateral and bilateral groups)(P>0.05).4.Visceral pain threshold in ratsPain thresholds were significantly lower in the model group compared with the null group(P<0.05);pain thresholds were significantly higher in the unilateral and bilateral groups compared with the model group P<0.05);No significant difference between the unilateral and bilateral groups(P<0.05).5.Changes of colonic slow wave frequency in ratsColonic slow wave frequency increased in the model group compared with the null group(P<0.05);slow wave frequency decreased in the unilateral group compared with the model group and also decreased in the bilateral group(P<0.05);there was no statistical significance between the unilateral and bilateral groups(P<0.05);No significant difference between the unilateral and bilateral groups(P<0.05).6.Enzyme-linked immunosorbent assay(ELISA)was used to detect the changes of cAMP and cGMP in serumCompared to the blank group,serum cAMP and cGMP levels were higher in the model group and the difference was statistically significant(P<0.05);compared to the model group,serum cAMP and cGMP levels were lower in the unilateral group and the difference was statistically significant(P<0.05);compared to the model group,serum cAMP and cGMP levels were significantly lower in the bilateral group and the difference was statistically significant(P<0.05);compared to the model group,serum cAMP and cGMP levels were significantly lower in the bilateral group and the difference was statistically significant(P<0.05).The difference was statistically significant(P<0.05);No significant difference between the unilateral and bilateral groups(P>0.05).Conclusion:(1)Both unilateral and bilateral electroacupuncture can effectively regulate the autonomic nerve balance of IBS-D rats,and significantly improve visceral pain sensitivity and colonic motor function;(2)There was no significant difference in the improvement of autonomic nerve balance,visceral hyperalgesia and colonic motor function in IBS-D rats,suggesting that acupuncture at unilateral acupoints can also be used for lighter patients. |
