To Explore The Mechanism And Curative Effect Of Shaoyao Liquorice Decoction In Treating Anal Fissure Based On Network Pharmacology

Objective:To explore the curative effect of Shaoyao Liquorice Decoction in treating patients with anal fissure,and to preliminarily explore its possible mechanism of action with the help of network pharmacology.Methods:1.Clinical efficacy observation Into hospital between January 2021 and December 2021,guangxi university first affiliated hospital of traditional Chinese medicine outpatient service,clear diagnosis of 40 patients with stage I anal fissure,randomized controlled trial of research Gui method,were randomly divided into control group and experimental group and control group given strong medicine hemorrhoid smoked lotion hip bath,anal outside ointment besmear,beautiful sheen sanzuotong embolism anus every time 1 grain,2 times a day,Treatment for 7days;On the basis of the control group,the experimental group was given oral Peony liquorice decoction,250 ml each time,twice a day,the course of treatment was the same as the control group.Gender,age,pain score and bleeding score were recorded.SPSS 23.0 was used for statistical analysis.2.Network pharmacology The main chemical components and targets of Shaoyaogao Decoction were obtained by TCMSP database,and the sexual components of Traditional Chinese medicine were screened according to two attribute values(bioavailability OB≥30%,drug-like DL≥0.18%).The main targets of anal fissure were obtained through Gencards,OMIM and Drug Bank databases,and protein interaction was analyzed using String platform to construct protein interaction network and the results were imported into Cytoscape3.7.2software to screen Hub nodes.Metascape platform was used to analyze "drug-compound-target" and its biological processes and pathways.Results:1.Clinical efficacy observation There was no difference in gender and age at baseline between the two groups(P > 0.05).Before treatment,there were no significant differences in bleeding score and pain score between the two groups(P > 0.05).After treatment,the bleeding score and pain score in experimental group were lower than those in control group(P < 0.05).2.Network pharmacology1)Effective active compounds and targets of Shaoyaogancao Decoction: 102 effective active compounds meeting the requirements of oral availability and drug-like characteristics were screened by TCMSP database,and 225 effective active compound targets of Shaoyaogancao Decoction were screened.2)Target proteins of anal fissure diseases: 1875 target proteins related to anal fissure diseases were screened by Gencards,OMIM and Drug Bank databases.3)Potential key targets of Shaoyao Liquorice Decoction in the treatment of anal fissure: 83 targets shared by Shaoyao Liquorice Decoction and anal fissure were obtained through Venny platform,which are potential key targets of Shaoyao Liquorice Decoction in the treatment of anal fissure.4)Construction of the "drug-compound target-disease" network of Shaoyao Liquorice Decoction for treating anal fissure: Based on Cytoscape3.7.2,a "drug-compact-target-disease" network of Shaoyao Liquorice Decoction for treating anal fissure was constructed.The network consists of 176 nodes and716 edges,including 1 disease node,2 drug nodes,102 active compound nodes and 83 target nodes.5)Interaction network of intersection target proteins:The interaction relationship between key target proteins obtained through STRING platform is visualized with Cytoscape3.7.2 software.The targets that play a key role in this network include IL6,AKT1,TNF,TP53,IL1 B,CASP3,ESR1,PTGS2,MAPK3 and STAT3,and obtain Hub nodes through Cytoscape3.7.2 software.The results showed that AKT1,TNF,IL6,TP53,STAT3,PTGS2,CASP3,MMP9,IL1 B and CXCL8 were closely related.6)Enrichment analysis results: Through the application of Metascape platform,97 items of molecular function,36 items of cell composition and1411 items of biological process were obtained,mainly involving cell reaction to nitrogen compounds,membrane raft,cytokine receptor binding,etc.Most of the targets are concentrated in the cancer pathway,the role of age-rage signaling pathway in diabetes complications,fluid shear stress and atherosclerosis,chemical carcinogen-receptor activation,inflammatory bowel disease,HIF-1 signaling pathway,relaxin signaling pathway,transcription disorders in cancer and other 120 signaling pathways.Conclusions:1.Shaoyao Liquorice Decoction has significant curative effect in the treatment of stage I anal fissure.2.Shaoyao Liquorice Decoction has the effect of multiple pathways and multiple targets in the treatment of anal fissure,which may be achieved by regulating 83 targets including AKT1,TNF,IL6,TP53,STAT3 and PTGS2 and their signal pathways in the course of anal fissure and affecting their related inflammatory factors.
