Chronic Bronchitis(CB)is a Chronic nonspecific inflammation of the mucous membranes of the trachea and bronchus and surrounding tissues.Traditional Chinese medicine(TCM)has rich experience in the treatment of chronic diseases,but different TCM have different drug use ideas and prescriptions for the treatment of the same diseases,and the drug use mechanism in the potential aspects of active ingredients,gene targets,and action pathways is still unclear.In this paper,chronic bronchitis was taken as an example to analyze the core prescription medication mechanism of TCM in the treatment of chronic bronchitis,explore the potential medication rule of TCM in the treatment of chronic bronchitis,and provide theoretical basis for clinical medication and new drug development.This study is divided into two parts.The first part: Based on the data of TCM outpatient service for chronic bronchitis treatment in a third class A hospital in Zhejiang Province in recent 6 years,multiple significant related drug groups and potential prescriptions were obtained through Apriori association analysis and systematic clustering,and the core prescriptions were extracted by comprehensive analysis.The second part: Firstly,the mechanism of action of the core prescription is analyzed by network pharmacology.Prescription active ingredients and gene targets were obtained from TCMSP database,chronic artery disease targets were obtained from GeneCard database,intersection targets were calculated,and imported into String database to construct PPI network diagram.Cytoscape software was used to construct topology network and screen core active ingredients and gene targets.Then Metascape database was used to analyze the GO and KEGG enrichment pathways.Finally,the mechanism of the core prescription was compared with the existing famous traditional Chinese medicine to explore the law.The results are as follows:(1)Data mining for outpatient data: Multiple different significantly related drug groups were obtained through Apriori association analysis,and 5 groups of new prescriptions were obtained through cluster analysis.After comprehensive analysis,the core prescriptions were obtained: chuanxiong,glycyrrhiza,platycodon grandiflorum,magnolia officinalis,perilla seed,terminalia chebula.(2)Pharmacological analysis of the core prescription network: the prescription contained 130 active ingredients for treating diseases,mainly including: luteolin,quercetin,etc.,134 genes that treat diseases,including ATK1,MAPK3 and other core target genes,mainly act on MAPK,PI3K-Akt,IL-17 and other signaling pathways,and reduce inflammation by inhibiting the expression of inflammatory factors and cell proliferation.(3)The pharmacological mechanisms of different core prescriptions were compared and analyzed: luteolin,quercetin and kaempferol were the main active ingredients of the three core prescriptions acting on the slow branches,mainly acting on MAPK3,IL-6 and EGFR,and the core targets were mainly concentrated on MAPK,PI3K-Akt and HIF-1 signaling pathway.The drug mechanism is mainly to inhibit the division,proliferation and migration of inflammatory cells,and reduce respiratory tract and airway inflammation to achieve the purpose of treatment.At the same time,the differences of individual core active ingredients,gene targets and pathways also reflect the unique use of famous old TCM in treating chronic bronchitis based on syndrome differentiation. |
