Qinlan oral liquid is composed of Lonicera japonica Thunb.,Forsythia suspensa(Thunb.)Vahl,Isatis indigotica Fort.and Scutellaria baicalensis Georgi.It has the effect of relieve fever and detoxification.It is mainly used to treat acute upper respiratory tract infection.Up to now,few studies have been reported on the pharmacodynamic material basis of Qinlan oral liquid.research.In order to explore the pharmacodynamic material basis and action mechanism of Qinlan oral liquid,provide scientific basis for follow-up clinical application,quality control and new drug research,we used UPLC-QTOF-MSE to identified chemical ingredients of Qinlan oral liquid in vitro and in vivo by Masslynx and references.In order to ensure the target organ of compound Qinlan oral liquid,we carried on the tissue distribution experiment.A total of 66 compounds were identified in Qinlan oral liquid,there are 13 flavonoids,9 organic acids,10 iridoids,9 phenylethanol glycosides,5 alkaloids,2 terpenoids,1 phenylpropanoid,2 lignans,12 others and 3 unknown compounds.Through serum pharmacochemistry experiment,12 compounds were found among the blood transitional components,including 3 prototype components and 9 metabolites.Then through tissue distribution experiment,the target organs of Qinlan oral liquid were liver,lung and kidney.Then the action mechanism of Qinlan oral liquid containing cold,fever,cough,sore throat was predicted through network pharmacology.The main action pathways involve 6 pathways,and the action targets are closely related to inflammation and immune response.To determine whether the network pharmacology results are accurate,We did the biological verification of cough symptoms in clinical,Firstly,we established the post infection cough(PIC)model in rats,and then the serum and tissue samples of rats were detected by ELISA,and we observed the pathological changes of lung tissue and the expression of PCR gene in lung tissue.The biological verification results indicated that after treatment of Qinlan oral liquid,cough can be effectively suppressed by reducing the expression of inflammatory factors.
