| Objective: By observing the effect of Tiaowei Shengqing Decoction on diabetic factor in model of type 2 diabetic rats,and the effect of Tumor necrosis factor-α(TNF-α)、Ibterleukin-1β(1L-1β)、Interferon-γ(INF-γ).Methods:Sixty healthy SD rats(male)were selected for adaptive feeding for one week,and randomly divided into 6 experimental groups,10 rats in each group were normal control group,T2 DM model group,metformin treatment group,Tiaowei Shengqing Decoction high-dose,medium-dose and low-dose groups respectively.In addition to normal group fed with conventional diet,the other 5 groups were fed with high-sugar and high-fat diet for 28 days.After 28 days,the 5 groups of rats were performed induction modeling by one-time intraperitoneal injection of streptozotocin-1(STZ)solution at a dose of 40mg·kg-1.After modeling,the patients were given intragastric administration for 8 weeks,and the changes of body weight and fasting blood glucose(FPG)and 2 hours postprandial blood glucose(2h PG)were recorded every 7 days.Oral glucose tolerance test(OGTT)was performed on day 35 of administration.Tiaowei Shengqing Decoction group was as follows:17.18g/kg;Medium dose group: 8.59g/kg;Low dose group: 1.72g/kg intragastric administration,control group metformin hydrochloride dose 250mg/kg.After the last administration,the rats were sacrificed for abdominal aorta blood sampling and liver tissue sampling.Testing the serum of the rats,Fasting insulin(FINS),alanine aminotransferase(ALT),aspartate aminotransferase(AST),total cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL-C),low density lipoprotein(LDL-C)and tumor necrosis factor-α(TNF-)were determined by ELISAα),interleukin-1β(1L-1β)and interferon-γ(INF-γ)were determined.The liver tissue was observed by HE staining.Results:Compared with normal group,body weight,FBG,2h PG,OGTT,FINS,TC,TG,ALT,AST and LDL-C contents in model group were significantly increased,while HDL-C was decreased(P < 0.05).HE staining results: Liver steatosis was increased in model group.After treatment,compared with model group,body weight,FBG,2h PG,OGTT,FINS,LDL-C,TC and TG of Tiaowei Shengqing Decoction high-dose,medium-dose and low-dose groups were improved(P < 0.05),while ALT and AST were significantly improved(P < 0.01).The inflammatory factors TNF-α,1L-1β and INF-γ of Tiaowei Shengqing Decoction were significantly controlled(P <0.01).HE staining results: Compared with model group,liver cell structure in Tiaowei Shengqing Decoction high-dose,medium-dose and low-dose groups was improved to varying degrees,steatosis was reduced to varying degrees,and inflammatory cell infiltration and structure were alleviated.Conclusion: Tiaowei Shengqing Decoction can effectively improve body weight and glucose metabolism index of T2 DM rats,relieve insulin resistance,inflammatory response and lipid metabolism disorder,and protect liver tissue and liver function of rats.It can reduce the levels of TNF-α,1L-1β and INF-γ in serum,suggesting that Tiaowei Shengqing decoction can treat T2 DM by inhibiting the expression of inflammatory factors. |
