ObjectiveObjective to analyze the anti-diabetic mechanism of chitosan oligosaccharides,and its regulatory effect on immune function and inflammatory factors release in rats,so as to provide reference for clinical treatment of type 2diabetes.Methods1.Twelve of the 64 SPF Kunming mice were randomly selected as the control group.The rest of the mice were treated with high-fat diet combined with low-dose intraperitoneal injection of streptozotocin(STZ)to establish the model of type 2 diabetes.48 mice were successfully established.They were randomly divided into model group,low-dose group,medium dose group and high-dose group,with 12 mice in each group.The model group,low-dose group,medium dose group and high-dose group were given 0.9%normal saline,100 mg·kg-1,200 mg·kg-1and 400 mg·kg-1chitosan oligosaccharides by gavage respectively for 30 days.The mice were killed 2 hours after the last administration.2.Fasting blood glucose level of mice was detected by tail vein blood sampling,OGTT test was conducted,FINS of mice was detected by automatic chemiluminescence analyzer,and HOMA-IR was calculated.3.TC and TG in serum of mice were detected by automatic biochemical analyzer,HDL-C and LDL-C were detected by selective precipitation method.4.The expression levels of CD4+and CD8+in mice were detected by flow cytometry,and the ratio of CD4+/CD8+was calculated.5.The expression levels of TNF-α,CRP,IL-6,IL-1βand MCP-1 in mice were detected by ELISA.Results1.Compared with the control group,the FBG level of the model group and low,medium and high dose group mice increased significantly(P<0.05),and with the increase of the treatment dose,the FBG level decreased(P<0.05),the OGTT results of the model group the low,medium,and high dose groups at different observation time were significantly higher than those of the control group(P<0.05),and with the increase of the treatment dose,the OGTT level was decreased(P<0.05),the indexes of Hb A1c,FINS and HOMA-IR in the in the model group and low,medium and high dose groups were significantly increased(P<0.05),and the indexes in the low,medium and high dose groups was lower than that in the model group(P<0.05).With the increase of treatment dose,the expression of Hb A1c,FINS and HOMA-IR gradually decreased(P<0.05).2.The lipid metabolism indexes of the model group and the low,medium and high dose groups were significantly different from those of the control group(P<0.05),TC,TG and LDL-C indexes of the low,medium and high dose groups were significantly lower than those of the model group(P<0.05),and HDL-C was higher than that of the model group(P<0.05).3.The thymus and spleen index and CD4+of the model group and the low and medium dose groups were significantly lower than those of the control group(P<0.05),while those of the low,medium and high dose groups were higher than the model group(P<0.05),and there was no significant difference between the five groups(P>0.05).4.TNF-α,CRP,IL-6,IL-1βand MCP-1 in the model group were higher than those in the control group(P<0.05),the inflammatory factors in different dose groups were significantly lower than those in the model group(P<0.05).ConclusionsChitosan oligosaccharide can play a good role in the regulation of glucose and lipid in diabetic mice,improve the immune function of mice and inhibit the release of inflammatory factors.
