| Objective:By observing the effect of Tiaowei Shengqing Decoction on the levels of blood sugar,blood lipids and insulin in model rats with high-fat and high-sugar diet combined with streptozotocin-induced glucose regulation impairment,and the effect of Leptin(LEP)、Tumor Necrosis factor alpha,(TNF-α)、Interleukin 6(IL-1),and liver histopathological changes in rats with impaired glucose regulation.To investigate the effects of Tiaowei Shengqing Decoction in IGR model rats,and to provide an experimental basis for the clinical application of Tiaowei Shengqing Decoction in the treatment of impaired glucose regulation.Methods:In the experiment,60 healthy 6-week-old Wistar rats were randomly divided into 6 groups,namely the normal control group,the IGR model group,the Tiaowei Shengqing Decoction low-dose group,the Tiaowei Shengqing Decoction medium-dose group,and the Tiaowei Shengqing Decoction high-dose group,metformin positive control group.After one week of acclimatization,five groups of rats,except for the normal control group,were continuously fed with high-fat and high-sugar diets.Four weeks later,the impaired glucose regulation rat model was induced by a single intraperitoneal injection of streptozotocin 10mg·kg-1in five groups of rats except for the blank control group.After successful induction of the model,four treatment groups of rats were subjected to drug intervention by gavage for 8 weeks.Meanwhile,rats in normal control group and IGR model group were gavaged with distilled water 2ml/(kg·d).After successful modeling,fasting blood glucose and 2-hour postprandial blood glucose were measured in all rats,and the body weight of rats was measured and recorded at weekly intervals.After the final drug intervention,2 hours later,the 2-hour postprandial glucose was measured in all rats,fasting blood was collected from the tail-tip vein of all rats after 12 hours of fasting without water to measure fasting blood glucose,and the body weight of all rats was weighed after 2 hours of water fasting.At the end of the collection and weighing,all rats were executed,their abdominal aortic blood and liver tissue were taken,and the specimens were processed using the appropriate methods.Measurement of changes in body weight of rats after therapeutic drug intervention.Changes in blood glucose levels in rats after therapeutic drug intervention were measured using a glucometer.The levels of the corresponding cytokines(LEP、TNF-α、IL-6)in rat serum were measured using ELISA assays.Measurement of four lipids(TC、TG、LDL、HDL)in the serum of all rats.The levels of insulin in the serum of all rats were measured.Pathological changes in liver tissue were observed under light microscopy using the HE staining method.Results:1.The results of body weight changes in rats showed that the body weights of rats in the IGR model group,the Tiaowei Shengqing Decoction low-dose group,the Tiaowei Shengqing Decoction medium-dose group,and the Tiaowei Shengqing Decoction high-dose group and the metformin-positive control group were significantly higher than those of the normal control group after feeding with high-fat and high-sugar diets and injection of STZ.After 4 weeks of therapeutic drug intervention,the rats in the medium and high dose groups of Tuning Stomach Ascending Soup showed slower weight gain than the IGR model group(P<0.05).At the end of the experiment,compared with the IGR model group,the body weight of rats treated with the low,medium and high doses of Tiaowei Shengqing Decoction and metformin-positive control group was significantly reduced(P<0.05).2.The results showed that the fasting blood glucose and 2-hour postprandial blood glucose of all the modeled rats increased significantly after the successful modeling compared with the normal control group(P<0.05).After 8 weeks of therapeutic drug intervention,the FPG and2h PG of the rats in the low,medium and high doses of Tiaowei Shengqing Decoction and metformin groups were reduced to different degrees compared to the IGR model group(P<0.05).3.ELISA results showed that,compared to normal controls,The levels of TC,TG and LDL were significantly increased in the IGR model group rats(P<0.05),Significantly lower HDL levels(P<0.05),Compared with the IGR model group,after treatment with low,medium and high doses of Tiaowei Shengqing Decoction and metformin drugs,The serum levels of TC,TG and LDL in rats were reduced to different degrees(P<0.05),HDL levels were elevated to varying degrees(P<0.05),Compared with normal controls,Fins levels were significantly higher in the IGR model rats(P<0.05),Compared with the IGR model group,after treatment with low,medium and high doses of Tiaowei Shengqing Decoction and metformin drugs,Fins levels in rats were reduced to different degrees(P<0.05),Compared with the normal control group,the serum levels of LEP,TNF-αand IL-6 were significantly higher in the IGR model rats(P<0.01),Compared with the IGR model group,the serum levels of LEP,TNF-αand IL-6 were significantly lower in the metformin group and the medium and high dose groups of Tiaowei Shengqing Decoction(P<0.01).4.HE staining showed that compared with the normal group,the hepatocytes in the IGR model group showed significant necrosis and a large number of fat vacuoles were found,accompanied by a large number of inflammatory cells infiltration(P<0.05).Compared with the IGR model group,hepatocyte necrosis was significantly reduced and the number of fat vacuoles was decreased in the rats treated with low and medium doses of Tiaowei Shengqing Decoction and metformin,but inflammatory cell infiltration was still found by staining results(P<0.05).In contrast,the staining results of the high-dose group of Tiaowei Shengqing Decoction revealed basically intact liver cells,slight fat vacuoles and inflammatory cell infiltration,no necrotic hepatocytes,normal arrangement of hepatic cords,and obvious delineation of liver lobules(P<0.05).Conclusion:Tiaowei Shengqing Decoction can effectively reduce the body weight and blood glucose index of rats with impaired glucose regulation,effectively regulate glucolipid metabolism,improve insulin resistance,effectively reduce fatty degeneration and deposition of hepatocytes,and protect liver function.Effectively alleviating the chronic inflammatory state in rats with impaired glucose regulation and reducing the levels of inflammatory factors LEP,TNF-αand IL-6 in serum.And it is speculated that the inhibition of inflammatory factor expression is one of the mechanisms of action of Tiaowei Shengqing Decoction for the treatment of impaired glucose regulation. |
