Effect Of Wenhua Juanbi Recipe On MiRNA-146a Regulating And Controlling DNA Methylation In Peripheral Blood Mononuclear Cell Of Patients With Rheumatoid Arthritis | | Posted on:2019-12-10 | Degree:Master | Type:Thesis | | Country:China | Candidate:A Q Wang | Full Text:PDF | | GTID:2544305450951429 | Subject:Traditional Chinese Medicine | | Abstract/Summary: | PDF Full Text Request | | Objective To study the effect of Wenhua Juanbi recipe(WJR)on the regulation of DNA methyltransferase DNA methyltransferasone(DNMTs)in peripheral blood mononuclear cells(PBMCs)from patients with active rheumatoid arthritis(RA),and to elucidate the molecular mechanism of DNA methyltransferase DNA methyltransferasestin(DNMTs)in treatment of RA.Methods Nine patients with active RA with cold and heat disorder and phlegm stasis obstruction were randomly divided into three groups: RA group was transfected with antisense miRNA-146 a inhibitor,and 9 healthy subjects were divided into 3 groups.They were normal group,normal transfection miRNA-146 a group(miRNA-146 a group,normal transfection miRNA-146 a + Chinese medicine group,3 cases in each group).In vitro cell culture,transfection and use of PBMC were carried out.Real-time quantitative PCR assay was used to detect the expression of PBMC miRNA-146 a..Results:(1)Compared with the normal control group,the expression of PBMC miRNA-146 a in active RA patients was increased(P < 0.05)and the expression of DNMTs was decreased(P < 0.05).(2)Compared with the normal control group,the expression of PBMC miRNA-146 a in the normal transfected miRNA-146 a group increased(P<0.05),and the expression of DNMTs decreased(P <0.01).(3)Compared with RA group,the expression of miRNA-146 a in antisense miRNA-146 a was decreased(P < 0.05)and the expression of DNMTs was increased(P < 0.05).Compared with the model group,the expression of PBMC miRNA-146 a in RA group was significantly lower than that in the model group(P < 0.05),the expression of DNMTs increased(P < 0.05).(4)Compared with RA group,the expression of PBMC miRNA-146 a in RA + TCM group was significantly lower than that in RA group(P < 0.05).(5)Compared with the normal control group,the expression of miRNA-146 a PBMC miRNA-146 a decreased(P < 0.05)and the expression of DNMTs did not change significantly(P < 0.05).Conclusion:(1)The abnormal expression of PBMC miRNA-146 a in active RA patients and healthy controls suggests that miRNA-146 a may be involved in the pathogenesis of RA.(2)miRNA-146 a may have a regulatory effect on DNMTs(3)Wenhua Juanbi prescription may increase the expression of miRNA-146 a and decrease the expression of DNMTs.It may affect the regulation of DNA methylation by miRNA-146 a,which is one of the mechanisms of its treatment of RA... | | Keywords/Search Tags: | Wenhua Juanbi prescription, (WJR), miRNA-146a, DNA methyltransferase in rheumatoid arthritis(DNMT1、DNMT3a、DNMT3b), Cell culture, transfection PCR, PCR | PDF Full Text Request | Related items |
