Objective:To study the therapeutic effect of Shenlijia Granule on chronic heart failure rats and the effect of Shenlijia Granule on MCP-1.Methods:Selected 15 rats as blank group randomly from 135 healthy Wistar male rats.The rest rats were intraperitoneally injected with Adriamycin for CHF animal model.Successful model rats were randomly divided into model group,traditional Chinese medicine group(Qiliqiangxin group),western medicine group(Digaoxin group),Shenlijia Granule low,medium and high dose group.There were 15 rats in each group with normal saline and corresponding suspension for 28 days.Blank group does not take measures.Observe the general state of the rat and removed the rat heart tissue.Using experimental methods such as cardiac ultrasound,hemodynamics,histomorphology,immunohistochemistry,and ELISA to analyze the experimental results.Results:1.CHF model was duplicated successfully.2.Shenlijia granules can improve the general state of heart failure rats and improve the quality of life of heart failure rats.3.Results of rat echocardiography:The LVIDs and LVIDd in the model group were significantly higher than those in the blank group,and the FS and EF were significantly lower.Compared with the model group,the LVIDs and LVIDd in all treatment groups were reduced to varying degrees,and FS and EF were increased to varying degrees.P<0.01.4.Heart function hemodynamics results in rats:The LVEDP in the model group were significantly higher than those in the blank group,and the LVSP、+dp/dtmax and-dp/dtmax were significantly lower.Compared with the model group,the LVEDP in all treatment groups were reduced to varying degrees,and the LVSP、+dp/dtmax and-dp/dtmax were increased to varying degrees.P<0.01.5.Pathology and transmission electron microscopy studies show that shenlijia granules can improve general and ultrastructure of myocardial in heart failure rats.6.Results of immunohistochemistry and ELISA:Compared with the blank group,the content of MCP-1 in the model group were decreased significantly.Compared with the model group,the content of MCP-1 in each treatment group were increased differently.7.The overall analysis of the experimental results show that Shenlijia granules in the medium dose group have the best therapeutic effect.Conclusion:1.The Shenlijia granules can significantly improve echocardiography and hemodynamics of heart function in CHF rats.2.Shenlijia granules medium dose group has better treatment effect than other treatment groups.3.Shenlijia granules can regulate the expression of MCP-1 during CHF.
