Objective:To study the action mechanism of Shenlijia particle regulating the myocardial TGF β-3 expression of rats with chronic heart failure(CHF)by the technology of micRNA chip and Network Database Comparison Analysis,and provide scientific basis for further study of Shenlijia particle preventing CHF by searching the target MicRNA.Methods:Healthy male Wistar rats were intraperitoneally injected with Adriamycin and CHF animal model was duplicated.Echocardiography,Hemodynamics,Histomorphology,Immunohistochemistry,ELISA,micRNA chip,QPCR and the technology of Network Database Comparison Analysis were applied to observe Shenlijia’s impact on general condition,cardiac function,structure of myocardial tissues,TGF β-3 and other indicators related to micRNA,as well as correlation analysis.Results:(1)CHF model was duplicated successfully;(2)Shenlijia particle can effectively improve the general condition and survival quality of the CHF rats;(3)Echocardiography and hemodynamics show that Shenlijia particle can dramatically improve the cardiac function in heart failure rats;(4)Pathology and Transmission Electron Microscopy(SEM)show that Shenlijia particle can improve the general structure and ultrastructure of CHF rats,and it can improve myocardial fibrosis and protect the integrity of cardiomyocytes as well as structure of organelle;(5)ELISA,Immunohistochemistry and QPCR show that Shenlijia particle benignly regulating the expression of TGF β-3 and the TGF β-3 difference expression between groups exists;(6)Immunohistochemistry shows that Shenlijia particle benignly regulating the expression of COL Ⅰ/Ⅲ and improve myocardial fibrosis;(7)All the Results shows that the overall effect of middle-dosage group is optimal;(8)According to the research of Network Database Comparison:Variance Analysis→ Trend Analysis→microRNA Target Prediction→GO Analysis→Pathway Analysis,we got three micRNA related to this research(micRNA-361-3p,micRNA-497-5p and micRNA-500-3p)which are confirmed to be related to TGF β-3 expression,TGFβ-3—Smad signal transduction pathway and Shenlijia particle preventing CHF.QPCR experimental study certifies that the difference expression between groups exists.Conclusion:Shenlijia particle can effectively prevent CHF,improve the myocardial fibers and reverse the ventricular remodeling,improve the cardiac function of CHF rats.Shenlijia particle benignly regulate the expression of TGF β-3 and many kinds of micRNA,particularly for micRNA-361-3p,micRNA-497-5p and micRNA-500-3p.GO Analysis and Pathway Analysis of Network Database show that micRNA-361-3p,micRNA-497-5p and micRNA-500-3p are related to TGF β-3 expression,TGF β-3—Smad signal transduction pathway and Shenlijia particle preventing CHF.It is concluded that one of the possible mechanisms of improving the myocardial fibers and reversing the ventricular remodeling is regulating the TGF β-3 expression by adjusting the expression of micRNA-361-3p,micRNA-497-5p and micRNA-500-3p,thus influencing the TGF β-3—Smad path. |
